Cold-Pressed Nigella Sativa Oil Standardized to 3% Thymoquinone Potentiates Omega-3 Protection against Obesity-Induced Oxidative Stress, Inflammation, and Markers of Insulin Resistance Accompanied with Conversion of White to Beige Fat in Mice

Abstract: Excessive lipid accumulation in white adipose tissue (WAT) results in adipocyte hypertrophy and chronic low-grade inflammation, which is the major cause of obesity-associated insulin resistance and consequent metabolic disease. The development of beige adipocytes in WAT (browning of WAT) increases energy expenditure and has been considered as a novel strategy to counteract obesity. Thymoquinone (TQ) is the main bioactive quinone derived from the plant Nigella Sativa and has antioxidative and anti-inflammatory … Show more

“…An emerging interest has been focused on the bioactive compounds that mitigate the progression of NAFLD. Polyphenols, such as thymoquinone [ 16 , 17 ], resveratrol [ 18 ], and pomegranate [ 19 ], have been reported to increase antioxidant capacities and HO-1 expression to regulate hepatic steatosis and inflammation [ 20 ]. Magnolol (MG) is the main bioactive compound derived from Magnolia officinalis , which has been shown to exert anti-inflammation, anti-oxidation, and hepatoprotective properties [ 21 , 22 ].…”

“…Recently, there has been growing interest in examining natural antioxidants found in dietary plants, which may have beneficial effects on metabolic disease. Previous studies have reported that curcumin, black seed oil, and resveratrol have beneficial effects on insulin sensitivity and on the inhibition of inflammation [25][26][27][28] Natural substances such as pomegranates may be beneficial towards combating obesity [29,30]. Pomegranate juice contains antioxidants such as polyphenols (0.2-1%), tannins, punicic acid, and ascorbic acid [31,32].…”

“…There are a number of naturally occurring substances that are capable of upregulating HO-1. These include Thymoquine (black seed oil) in combination with Omega-3 that was shown to protect against obesity induced oxidative stress, improve insulin sensitivity and convert white fat to beige fat in a murine model of obesity [ 157 ]. In addition, pomegranate seed oil upregulated HO-1, improved mitochondrial function, and attenuated hepatic steatosis/fibrosis in a murine model of obesity [ 158 ].…”
