Combination of CE‐MS and advanced chemometric methods for high‐throughput metabolic profiling

Abstract: In this work, an untargeted approach based on capillary electrophoresis-mass spectrometry (CE-MS) in combination with multivariate data analyses is proposed as a high-throughput general methodology for metabolomic studies. First, total ion electropherograms (TIEs) were considered for exploratory and classification purposes by means of principal component analysis (PCA) and partial least squares discriminant analysis (PLS-DA). Then, multivariate curve resolution alternating least squares (MCR-ALS) was applied t… Show more

“…Then, the resulting data matrices were further reduced in their m/z mode dimension in 30 different m/z ranges ( m/z widths for reduction were 20, 50, and 100 m/z in the m/z ranges 40–400, 400–800, and 800–1250 m/z , respectively; Fig. B) .…”

“…This robustness is demonstrated by the fact that satisfactory results can also be obtained when dealing with lower resolution data. For instance, MCR-ALS has successfully overcome inherent difficulties of GC-MS and CE-MS data analysis, such as multiple MS signals for the same metabolite due to derivatization in GC-MS and large migration time shifts between samples in CE-MS [33,34].…”

“…Several groups consistently trust CE‐MS for metabolomic applications in a routine productivity environment with remarkable results . Notwithstanding its advantageous features, CE‐MS remains an oddity in the field of metabolomics when compared to the more widely used chromatography‐based approaches.…”
