Combination of Immunotherapy and Photo‐pyroptosis as Novel Anticancer Strategy

Abstract: Immunotherapy has made great progress in clinical cancer treatment in recent years, but its therapeutic efficacy is significantly limited by the lack of immunogenicity in the tumor microenvironment. Pyroptosis is a type of programmed cell death in which the dying cancer cells produce inflammatory cytokines to relieve the immuno-suppressive microenvironment and thus increase anti-tumor immunity. “…Similarly to apoptosis, pyroptosis is induced by ROS produced during PDT with additional systemic immune responses toward solid tumors. As enhancement of systemic immune responses is crucial in anticancer PDT treatment, new types of PSs with the ability to invoke pyroptosis rather than apoptosis have been introduced in this field 313,314 …”

“…As enhancement of systemic immune responses is crucial in anticancer PDT treatment, new types of PSs with the ability to invoke pyroptosis rather than apoptosis have been introduced in this field. 313,314 Contrary to ferroptosis, pyroptosis is initiated by participation of ROS-activating inflammasomes followed by triggering pyroptosis-specific inflammatory caspases which cause the cleavage of gasdermin-D. 315 With the following release and translocation of gasdermin-D N-terminal domain, pores are formed on the cell membrane, which leads to cell death processes such as cell swelling, membrane disruption, or immunostimulatory release of cellular content. 313,316 Importantly, the formation of pores on cell membranes is driven by released N-terminal domain binding with membrane phospholipids.…”

“…Innovations such as bionic nanoparticles, which synergize indocyanine green and decitabine to trigger pyroptosis, have shown to attenuate immunosuppression and bolster systemic immunity, effectively combating primary and metastatic tumors ( 115 ). Additionally, the generation of reactive oxygen species (ROS) during photodynamic therapy (PDT) has been identified as a potent inducer of pyroptosis, with new photosensitizers capable of triggering immunogenic cancer cell pyroptosis, broadening the horizon for PDT applications ( 116 , 117 ).…”

“…[18][19][20][21] It is known that in the ICD process, damage-associated molecular patterns (DAMPs) can induce the expression of calreticulin (CRT) and heat shock protein 90 (HSP 90), as well as secretion or release of high mobility group box 1 (HMGB1) and ATP. [4,16,[22][23][24][25][26] These antigen signals are captured and identified by dendritic cells and macrophages. After processing, it was presented to adaptive immune cells, thus activating the anti-tumor immune response of the body.…”
