“…Hence, the adventitious shoot buds of N. cadamba developed de novo and from the explant epidermal cells as a result of mitotic activity of the epidermal cells in response to appropriate hormone concentrations, but not from preexisting meristems (vascular elements). This emergence is similar to that of the sunflower (Sujatha et al 2012), but different from Centaurea ultreiae, in which meristematic cells arise from the original vascular elements (Mallon et al 2011).…”

“…As the RHA280 line that was used for most of the research described here was generally more responsive to shoot induction than the single genotype evaluated in that study, the RHA280 line may show an enhanced shoot production that is not typical for all sunflower lines. Genetic control of shoot organogenesis (Sarrafi et al 1996) as well as variation in regeneration among genotypes (Sujatha et al 2012b) has been previously reported for sunflower cotyledon tissues. Shoot formation was obtained from preconditioned leaves of all seven genotypes evaluated in the present study, although the efficiency differed considerably (Table 3), further validating a genetic influence on shoot formation in sunflower tissues.…”

“…More recently, genetic transformation of cotyledonary tissues using Agrobacterium has also been reported (Sujatha et al 2012a). Shoot formation in sunflower has primarily been obtained using the shoot apex (Paterson 1984), embryo axis or preferably cotyledon explants from seeds or seedlings (Power 1987;Knittel et al 1991;Sujatha et al 2012b). Shoot buds have been obtained from expanding leaves of cultured shoot apices following culture of the leaves on a medium containing benzylaminopurine (BA) (Konov et al 1998).…”

“…As a result of inefficiencies in sunflower regeneration, transgenic approaches for basic research and improvement of this important oilseed crop have been somewhat limited. Thus far, shoot organogenesis in sunflower has been reported using shoot tips (Paterson 1984), cotyledon tissues from mature embryos (Power 1987;Dhaka and Kothari 2002;Sujatha et al 2012) and germinated seedlings (Knittel et al 1991;Ceriani et al 1992;Baker et al 1999), immature embryos (Müller et al 2001), and leaf tissues (Konov et al 1998;Zhang and Finer 2015). Due to year-round availability and high-shooting response from mature embryos and seedling materials, these tissue sources are preferred over immature embryos for sunflower regeneration.…”

“…Due to year-round availability and high-shooting response from mature embryos and seedling materials, these tissue sources are preferred over immature embryos for sunflower regeneration. With continuous culture on cytokinin-containing medium, cotyledonary tissues from dry mature seeds were more responsive to shoot induction than cotyledons from germinated seeds (Power 1987;Sujatha et al 2012). The cytokinin 6-benzylaminopurine (BA) was used most often for studies on sunflower shoot production (Paterson 1984;Power 1987;Knittel et al 1991;Dhaka and Kothari 2002), as it was more effective than other cytokinins (Paterson 1984).…”
