Combined administration of cerebrolysin and donepezil induces plastic changes in prefrontal cortex in aged mice

Abstract: Cerebrolysin (Cbl) shows neurotrophic and neuroprotective properties while donepezil (Dnp) is a potent acetylcholinesterase (AChE) inhibitor, both drugs are prescribed for Alzheimer's disease (AD) treatment. Previous studies have shown that the Dnp and Cbl administered separately, modify dendritic morphology of neurons in the prefrontal cortex and hippocampus in senile rodents. Since the deficit of neurotrophic factor activity is implicated in the degeneration of cholinergic neurons of basal forebrain, a combi… Show more

“…Accumulated evidence from previous studies suggests that DON possesses neuroprotective properties in the suppression of neurodegeneration (15,36). Studies have reported that the neuroprotective effects of DON slow the progression of hippocampal atrophy in Alzheimer’s disease (37), protect cortical neurons in models of oxygen-glucose deprivation and glutamate-induced toxicity, protect against the effects of hippocampal mitochondrial dysfunction in transgenic mouse models (38), and increase the total dendritic length and spine density of neurons in aged mice (39). In addition, DON treatment has been shown to be effective in preserving presynaptic protein in the hippocampus and spinal cord in a tauopathy mouse model (40).…”

“…DON may induce its neuroprotective effect by activating the neurotrophin receptors in the hippocampus (41). In addition, it has been shown that DON protects neurons by upregulating nicotinic acetylcholine receptor subtypes to decrease the glutamate toxicity that is involved in a number of neuronal degenerative diseases (36,39,42). In this context, the recovery of synaptic protein munc-18 in the co-administration group may occur as a result of DON-induced neuroprotection against hippocampal neuronal impairment, leading to an altered synthesis of the synaptic proteins.…”

“…These results may suggest that dendritic spine atrophy could be interpreted as maladaptive plasticity. Several reports have demonstrated that aging‐induced dendritic length hypotrophy and spine loss are accompanied by cognitive deficits in tasks mediated by the hippocampus and the PFC (Acosta‐Peña et al, ; Alcantara‐Gonzalez et al, ; Duan et al, ; Juarez et al, ; Romero‐Curiel et al, ). In addition, reduced spinogenesis in the PFC‐, hippocampus‐pyramidal and NAcc‐medium spiny neurons has been reported in animal models of VD (Sanchez et al, ; Vega et al, ).…”

“…The Sholl analysis was originally described by Donald Arthur Sholl in 1953 in his publication describing dendritic organization in the neurons of cat cortices (Sholl, 1953). This program computes the number of intersections in concentric annuli as a function of increasing distance from the neuron soma and has been used for decades to study dendrite development and morphology under various conditions (Aizawa et al, 2004; Alcantara-Gonzalez et al, 2012; Chen et al, 2005; Chiang et al, 2011; Milosevic and Ristanovic, 2007; Ristanovic et al, 2006; Schoenen, 1982; Sholl, 1953; Torres-Garcia et al, 2012). Finally, neuroscientists and biomedical engineers have occasionally borrowed from fiber orientation analysis to apply Fast Fourier Transform (FFT) to analyze neurite alignment (Ayres et al, 2006; Ayres et al, 2008; Levitt et al, 2007; Sander and Barocas, 2009; Sivaguru et al, 2010; Voyiadjis et al, 2011).…”
