Combined autoradiography and histochemistry: the simultaneous detection of 6‐³H thymidine and acid phosphatase activity in cryostat sections

Abstract

SUMMARY A method is described for the simultaneous demonstration of 6‐3H thymidine incorporation and acid phosphatase activity in cryostat sections of mouse thymus. The method enables a comparison of mitosis and acid hydrolase activity to be made in the same tissue section. In 8‐week‐old mice acid phosphatase positive cells represent 1·23 ±0·06% of the total population and 8·4 ± 0·27% of the cells incorporate tritiated thymidine. Acid phosphatase activity can be used to estimate cell autolysis and death. The implications of the method in relation to tissue dynamics is discussed.

This procedure provides a more accurate picture of thymidine incorportion than that presented in our preliminary studies (Lewis et al, 1979). The thymidine incorporation figures reported here are generally lower than those previously published for several reasons, the thymidine dose is reduced from 4 to 3 pCi/g body wt, the emulsion is also thinner, Ilford L4 diluted 1 : 5 with double distilled water, and the exposure time is reduced.…”
