Combined effect of retinoic acid and calcium on the in vitro differentiation of human adipose-derived stem cells to adipocytes

Abstract: These results suggest that ATRA and calcium together have significant inhibitory effect on adipogenesis that can be helpful for finding new mechanisms to prevent or control the adipogenesis.

“…For chondrogenesis, the chondrogenic medium was used which contained the same complete medium and dexamethasone (100 nM), L-proline (40 µg/ml; Merck Millipore Corporation, Istanbul, Turkey), L-ascorbic acid 2-phosphate (50 µg/ml; Sigma-Aldrich, Germany), and TGF-β3 (10 ng/ml; BioLegend Ò , Istanbul-Turkey). For adipogenesis, the adipogenic medium contained dexamethasone (1 µM), indomethacin (100 µM), insulin (10 µg/mL), and 3-isobutyl-1-methylxanthine (IBMX; 0.5 mM) (All from Sigma-Aldrich Co. Ltd., Dorset, UK) [17,18].…”

“…Some animal studies show that xenograft transplantation of mesenchymal stem cells can reduce the immunomodulation [48] and efficacy of these cells [49]. However, the use of hADSCs has been usual in preclinical studies [5052] and its efficacy is proven. The findings of the present study also show that xenograft transplantation of hADSCs in the anal sphincterotomy model in rabbit improves sphincter function and tissue repair.…”

“…The sterile samples were kept in a tube including DMEM/F12, FBS 10%, and penicillin/streptomycin (P/S) 5%. Like our previous works (18,19), we used a protocol described by Dubois et al for the isolation of stem cells (20). Before extracting, the fat samples were heated to 37°C in a water bath.…”
