Combined effects of systemic parathyroid hormone (1–34) and locally delivered neutral self‐assembling peptide hydrogel in the treatment of periodontal defects: An experimental in vivo investigation

Abstract: Aim To evaluate in vivo combination therapy of systemic parathyroid hormone (PTH) and locally delivered neutral self‐assembling peptide (SAP) hydrogel for periodontal treatment. Materials and Methods Viability/proliferation of rat periodontal ligament cells in a neutral SAP nanofibre hydrogel (SPG‐178) was evaluated using WST‐1 assay. Periodontal defects were created mesially to the maxillary first molars in 40 Wistar rats. Defects were filled with 1.5% SPG‐178 or left unfilled. Animals received PTH (1‒34) or … Show more

“…Furthermore, the CT in P 11 -4-treated defects appeared healthier, with a high condensation of newly formed blood capillaries which promote wound healing ( Guo and DiPietro, 2010 ). Our results are consistent with the findings of others using different SAPs, who concluded that SAP nanofiber scaffolds accelerate wound healing and re-epithelialization ( Schneider et al, 2008 ; Meng et al, 2009 ; Yoshida et al, 2019 ).…”

“…For the observation of PCNA-, VEGF-, and Osx-positive cells, magnification was set at ×200, and a field of connective tissue (350 µm × 470 µm area) was randomly sampled in each section and submitted to quantitative analysis using an image analyzer (Image-Pro Plus 6.2, Media Cybernetics, Rockville, MD, USA), essentially as described previously [31]. On each specimen, analysis sites were compartmentalized into three areas (Root side, Bone side, and Middle area) to count PCNA-, VEGF-, or Osx-positive cells [24,32].…”

“…To assess the effect of different concentrations of M101 on EC metabolic activity, an AlamarBlue assay (Life Technologies, Saint-Aubin, France) was performed as described previously 27 . Twenty-four hours before the experiment, 2 × 10 5 EC were seeded in each well of a 24-well plate.…”
