Combined proteome and transcriptome analyses for the discovery of urinary biomarkers for urothelial carcinoma

Abstract: Background:Proteomic discovery of cancer biomarkers in body fluids is challenging because of their low abundance in a complex background. Altered gene expression in tumours may not reflect protein levels in body fluids. We have tested combining gene expression profiling of tumours with proteomic analysis of cancer cell line secretomes as a strategy to discover urinary biomarkers for bladder cancer. Methods:We used shotgun proteomics to identify proteins secreted by three bladder cancer cell lines. Secreted prot… Show more

“…In this context, urine could prove to be an important proximal fluid in providing biomarkers that could be tested noninvasively [2]. The choice of urine is particularly useful in the context of kidney and urinary system related diseases such as acute and chronic kidney diseases, diseases with either the urinary bladder or prostrate, and kidney transplantation [2][3][4][5][6][7][8][9]. Circulating peptides in urine are to be considered potentially useful as biomarkers as they are both abundant and easily accessible.…”

“…The recent introduction of new high-throughput technologies resulted in the discovery of many new substances, which may be detected in urine and used for a noninvasive detection as tumor markers [16,17]. Combination of individual markers facilitates higher sensitivity of tests [18].…”

“…5,19 However, the latest generation of experimental platforms shows significant promise in the field of urinary biomarker discovery, identifying protein-, DNA-and RNA-based biomarkers with potential clinical utility. 20,21 A number of studies are ongoing, although for the foreseeable future it is likely that these biomarkers will only reduce the frequency of cystoscopic surveillance and not replace cystoscopy altogether, especially for high-risk NMIBC. However, we are hopeful that the future will see the development of a multi-biomarker urinary test with utility for both the diagnosis and surveillance of UBC.…”
