Coming of Age: Cryo-Electron Tomography as a Versatile Tool to Generate High-Resolution Structures at Cellular/Biological Interfaces

Abstract: Over the last few years, cryo electron microscopy has become the most important method in structural biology. While 80% of deposited maps are from single particle analysis, electron tomography has grown to become the second most important method. In particular sub-tomogram averaging has matured as a method, delivering structures between 2 and 5 Å from complexes in cells as well as in vitro complexes. While this resolution range is not standard, novel developments point toward a promising future. Here, we provi… Show more

“…The crosstalk between different types of transporters unfortunately remains understudied. With the advancement in membrane protein reconstitution [ 166 , 167 ], protein structural analysis of cellular events at near atomic resolution by cryo-electron tomography [ 168 ], and biophysical methodology of multi-subunit macromolecules, the existing data of cell biology, biochemistry, and structural biology on P4-ATPases and ABC lipid transporters should promote interdisciplinary biochemical, biophysical and computational approaches to elucidate the cross-talk among different lipid transporters in regulating membranes and membrane-mediated cellular signaling.…”

“…Electroporation uses short and high-voltage electrical pulses to temporarily create small holes in membranes, increasing their permeability and allowing hydrophilic cargoes to diffuse into EVs [67]. This technique is one of the most commonly used for EV loading.…”

“…It is particularly well suited to investigate protein‐membrane systems. [66] CryoET has recently been used to image the impact of Aβ on lipid vesicles. [23] Different Aβ assembly forms have been incubated with large unilamellar lipid vesicles (LUVs) containing; PC, Cholesterol and GM1 (68 : 30 : 2 by weight).…”
