Comparative analysis of an mcr-4 Salmonella enterica subsp. enterica monophasic variant of human and animal origin

Abstract: The divergence in genomes, plasmids and gene variants demonstrated that there was not a unique mcr-4-positive, monophasic Salmonella lineage circulating in animals and causing gastroenteritis in humans in Italy. There was no horizontal transfer of the same plasmid among Salmonella strains of animal and human origin, but the mcr-4 gene and a fragment of the plasmid identified in the animal strain were mobilized by an IS1294 into a different ColE plasmid.

“…Moreover, all mcr genes except ( mcr-6 and mcr-9 ) have been detected in samples from China. Thus far, mcr-2 (Belgium and Spain), mcr-3 (Brazil, Denmark, France, Germany, Japan, Spain and Thailand), mcr-4 (Italy and Spain), mcr-5 (Colombia, Japan, Spain and Germany), and mcr-9 (US) [27,29,30,31,32,33,34,35,36,37].…”

“…The putative plasmid contigs were analyzed and re-assembled to build single circular plasmids by mapping reference complete plasmid genome. Additional sets of PCRs were performed to confirm the circular status of predicted plasmids as reported previously ( Carattoli et al., 2018 ; Xu et al., 2018 ). Genes were predicted with GeneMarkS™ and further annotated by BLASTP and BLASTN against NR databases and SwissProt.…”

“…In addition to bacterial traits, the host range of Salmonella 4,[5],12:i:- also plays a role in the success of this global pathogen, with outbreaks linked to food products derived from livestock 11 , 12 , 25 , 26 , particularly swine. For example, colistin resistance has been reported in Salmonella 4,[5],12:i:- isolates from pigs and humans in China, where there has been use of polymyxins as growth supplements in swine 14 , 18 , 27 . Further, mcr genes mediating colistin resistance have also been identified on plasmids with resistance genes to multiple antimicrobials, including third-generation cephalosporins (3GCs) 14 , 18 , 23 .…”
