Comparative analysis of iol clusters in Lactobacillus casei strains

Abstract: The present study was designed to expand genetic knowledge of myo-inositol (MI) metabolism in Lactobacillus casei. Twenty-four L. casei isolates of dairy origin were tested for the presence of iol cluster. PCR screening revealed eight strains encoded functions involved in MI utilization, of which one strain was able to use MI as carbon source. To gain a deeper understanding of the function of iol genes, four of the eight observed iol clusters were subjected to the full sequencing procedure. The results showed … Show more

“…So far, only some strains of L. casei have been found to metabolize MI with poor efficiency. One should note that even the presence of genes encoding enzymes for MI metabolism may not be sufficient to confer an ability to utilize MI (Zhang et al 2010; Vinay-Lara et al 2014). Moreover, we identified glgBCDAP-amyB g enes connected with glycogen metabolism ( LCAKO_2211 to LCAKO_2216 ) organized identically to operons of other Lactobacillus species (Goh and Klaenhammer 2014).…”

“…In many inositol-degrading bacteria, multiple paralogous genes annotated or shown to encode IDHs were identified ( 39 41 ), which, in the case of B. subtilis , for example, enable growth not only on MI but also on SI and DCI ( 42 , 43 ). In C. glutamicum , seven potential inositol oxidoreductases were annotated, three in cluster iol1 (IolG, IolW, and OxiB), three in cluster iol2 (OxiC, OxiD, and OxiE), and another one elsewhere in the genome (IdhA3) ( Fig.…”

“…It can also be metabolised by bacteria that live in soil, but it is not frequently used as an energy source in LAB [ 39 ]. So far, strains of L. casei are the only members of LAB that are capable of weakly metabolizing MI, but the presence of a MI metabolism cluster of genes is not a common feature of L. casei strains, and it does not necessarily mean that the strain carrying the cluster will use it as an energy source [ 40 ]. Previously, the presence of the complete MI utilisation operon was confirmed in the probiotic strain L. casei BL23 [ 39 ].…”
