Comparative aspects of murine proteinase 3

Abstract: The neutrophilic granule protein proteinase 3 (PR3) is the preferred target antigen of anti-neutrophil cytoplasmic antibodies (ANCA) found in the serum of patients with Wegener's granulomatosis, a systemic small-vessel vasculitis. Due to the lack of data concerning the murine homologue of human PR3, we assessed the neutrophil marker polypeptide PR3 in mice by generating a murine-specific PR3 antibody. In contrast to humans, peripheral blood leukocytes are not the main resource of murine PR3. Interestingly, we … Show more

“…None of the cells and organs expressed traceable quantities of a truncated isoform of PR3. Note, that breakdown products of PR3 are visible in the bone marrow lane, which was already described in Relle et al [11]. M: Protein ladder in kDa.…”

“…For the detection of murine PR3, harvested CMT 64/61, Meta 7, RAG, WEHI 274, and NIH-3T3 D4 as well as spleen, kidney, lung, and bone marrow cells were lysed and 30 mg of lysate was subjected to an 8% SDS-PAGE. Blotting with a rabbit anti-mouse PR3 peptide antibody was done as previously described [11].…”

“…There is, however, increasing evidence of major functional differences between the human and murine immune systems, indicating limited application of data obtained in studies employing murine cells or models ( 5 , 6 ). On one hand, the composition of the murine leukocyte repertoire differs from that of humans; in mice only 7–28% of peripheral blood leukocytes are PMNs in contrast to 35–70% in human blood ( 7 ). On the other hand, functional differences, such as the sequence of participating immunological cells after antigen challenge as well as the dose of antigen needed to initiate immunological reaction in delayed type hypersensitivity have been described ( 5 ).…”

“…The difficulties in developing animal models for PR3-ANCA vasculitis are in part due to important species differences with respect to expression and physicochemical properties of PR3 [37,38]. To circumvent these differences, current strategies aim to develop humanized experimental model systems.…”
