BackgroundAs a promising candidate for artificial enzymes, catalytically active nanomaterials show several advantages over natural enzymes, such as controlled synthesis at low cost, tunability of catalytic activities, and high stability under stringent conditions. Rod-shaped Au–Pt core/shell nanoparticles (Au@Pt NRs), prepared by Au nanorod-mediated growth, exhibit peroxidase-like activities and could serve as an inexpensive replacement for horseradish peroxidase, with potential applications in various bio-detections. The determination of measles virus is accomplished by a capture-enzyme-linked immunosorbent assay (ELISA) using Au@Pt NR-antigen conjugates.ResultsBased on the enhanced catalytic properties of this nanozyme probe, a linear response was observed up to 10 ng/mL measles IgM antibodies in human serum, which is 1000 times more sensitive than commercial ELISA.ConclusionsHence, these findings provide positive proof of concept for the potential of Au@Pt NR-antigen conjugates in the development of colorimetric biosensors that are simple, robust, and cost-effective.Electronic supplementary materialThe online version of this article (10.1186/s12951-018-0371-0) contains supplementary material, which is available to authorized users.

The morphologic classification was the very first method to differentiate the microorganisms based on microscopic observation. Around the year 1675, Antoni van Leeuwenhoek constructed the first microscopes, which were very simple instruments with soda lime glass lenses made by a melting process. He achieved already a 500-fold magnification and could differentiate the morphology of some microorganisms (such as cocci or bacilli (rods)). However, his work was hardly respected and received little attention at that time. Thus, he kept the secret of how he created the lenses, and for a long time no one was able to reconstruct his microscopes.Modern bacteriology/microbiology began in the nineteenth century with the development of high-performance microscopes using achromatic optical lenses, and pioneers like Ferdinand Cohn, Louis Pasteur, and Robert Koch are considered the founders of these disciplines.While the first taxonomy of microorganisms was related to morphologic properties, which are still anchored in the nomenclature (e.g., Streptococcus spp. or Rhodospirillum spp.), modern bacteriology discovered different metabolic processes that can be used to differentiate organisms and are more suitable to classify their taxonomy. Modern genetics, which began with the works of Gregor Johann Mendel in the mid-nineteenth century, opened new prospects, and in 1977 Carl Richard Woese published the most influential work of the phylogenetic taxonomy based on 16S RNA [1].Presently, species differentiation of the (pathogenic) organisms is performed by their morphologic (microscopy) and metabolic properties (culture-based) as well as genetically (NAT (nucleic acid amplification technique)-based). Thereby, microscopy and culture-based differentiation is still quite similar to those procedures performed in nineteenth century by Koch and coevals! Background. Enzyme-linked immunosorbent assay (ELISA) is the only serological method approved for blood screening in China. Automated chemiluminescence immunoassay (CLIA) and electrochemiluminescence immunoassay (ECLIA) had been used in clinical laboratories but not applied to screen HIV among blood donors. This study aimed to evaluate the performance of ELISA, CLIA, and ECLIA, focusing on the feasibility of CLIA/ECLIA for blood screening. Method. 1029 blood donations from 14 blood centers screened by ELISA were enrolled in the study. All plasma samples were tested by eight ELISA assays in 16 blood centers, followed by the detection of CLIA and ECLIA methods in the National Center for Clinical Laboratories (NCCL), further confirmed by nucleic acid testing (NAT) and Western blot (WB). Results. Of 1029 samples, 136 were confirmed as HIV positive. CLIA and ECLIA assay had similar sensitivities with ELISAs but showed higher specificity (CLIA: 99.1%, 885/893; ECLIA: 99.0%, 884/893), concordance rate (CLIA: 99.2%, 1021/1029; ECLIA: 99.1%, 1020/1029), and positive predictive value (PPV) (CLIA: 94.4%, 136/144; ECLIA: 93.8%, 136/145) than most of ELISA kits (>5 ELISAs) (P<0.05). Kappa values of CLIA (0.967) and ECLIA (0.963) were the highest among all the serologic assays. Among 451 samples with initial ELISA reactivity, 315 were negatives, of which 307 (97.5%) and 306 (97.1%) were detected as nonreactive by CLIA (8 nonspecific reactions) and ECLIA (9 nonspecific reactions), respectively. Conclusion. Compared with ELISA, CLIA and ECLIA are more specific and accurate in detecting HIV antibody/antigen and can keep more nonspecifically reactive donors detected by ELISA. CLIA and ECLIA can be used for the improvement of serological blood screening strategy to avoid the unnecessary loss of blood donors.
