Comparative growth of trichoderma strains in different nutritional sources, using bioscreen c automated system

Abstract: Trichoderma is one of the fungi genera that produce important metabolites for industry. The growth of these organisms is a consequence of the nutritional sources used as also of the physical conditions employed to cultivate them. In this work, the automated Bioscreen C system was used to evaluate the influence of different nutritional sources on the growth of Trichoderma strains (T. hamatum, T. harzianum, T. viride, and T. longibrachiatum) isolated from the soil in the Juréia-Itatins Ecological Station (JIES),… Show more

“…curves obtained. These improvements in the microtitre well format of the Bioscreen C have allowed the extension of the incubation time over which measurements can be made from 48 (Schnurer 1993;Meletiadis et al 2001aMeletiadis et al , 2001bRossi-Rodrigues et al 2009) to 72 h and then extending this further to 7þ d. The independence of the volume used also makes this technique appropriate for a number of applications. This is supported by the screening of PTS (18 concentrations) at different a w and temperature conditions which could be done over periods of up to 7 d. This also suggests that it may be possible to use these data sets obtained on filamentous and mycotoxigenic spoilage fungi to accurately model the efficacy of ecological factors and chemical control agents.…”

“…Spectrophotometric assay methods based on turbidimetric measurements have been used in a small number of mycological growth studies (Schnurer 1993;Rossi-Rodrigues et al 2009). These studies have been mainly in the medical field (Van Eldere et al 1996;Meletiadis et al 2001aMeletiadis et al , 2001bMeletiadis et al 2003) and used to develop in vitro antifungal susceptibility testing of conidium-forming fungi; this has provided rapid results that are expressed in Optical Density (O.D.)…”

“…Since fungi can grow in extreme environments, have large cell size and require simple culture conditions when grown on large scale, they are ideal candidates for the conversion of fatty acids to alkanes. [5,6] Some fungal species accumulate oil under certain cultivation conditions, which is an important advantage for the production of biofuels and industrial chemical feedstocks. [7,8] The selection of fast-growing productive fungal strains optimized for desired growing conditions is of fundamental importance for the production of high-value products.…”

“…[6,9] We used Trichoderma koningii, Penicillium janthinellum and their mixed culture in controlled batch culture studies to determine their ability to grow, oxidize and convert decanoic acid (DA), undecanoic acid (UDA), their mixture and UDAC potato dextrose broth (UDACPDB) into intracellular lipids during 24 days of incubation.…”
