Comparative study of Mg/Al- and Zn/Al-layered double hydroxide-perindopril erbumine nanocomposites for inhibition of angiotensin-converting enzyme

Abstract: Abstract:The intercalation of a drug active, perindopril, into Mg/Al-layered double hydroxide for the formation of a new nanocomposite, PMAE, was accomplished using a simple ion exchange technique. A relatively high loading percentage of perindopril of about 36.5% (w/w) indicates that intercalation of the active took place in the Mg/Al inorganic interlayer. Intercalation was further supported by Fourier transform infrared spectroscopy, and thermal analysis shows markedly enhanced thermal stability of the activ… Show more

“…14 Owing to the efficient drug-delivery feature of LDH nanocarriers, many pharmaceutically active compounds have been successfully intercalated into the interlayer gallery of LDH. Among them are the cardiovascular drugs fluvastatin and pravastatin; 15 anti-inflammatory drugs such as diclofenac 16 and fenbufen; 17 antihypertensive drugs like perindopril erbumine; 18,19 the antihistamine drug cetirizine hydrochloric acid; 20 and anticancer drugs such as cordycepin, which was intercalated into the gallery of magnesium/ aluminum (Mg/Al)-LDH. It was observed that the resulting nanohybrid had greater stability and a greater suppression effect on U937 cancer cell growth than free cordecypin 3 and methotrexate (MTX).…”

“…16,37 spatial orientation of INh into the interlayer gallery of Mg/al lDh Figure 2A shows the values for the long and short axes and thickness of INH, which were 10.5 Å , 7.2 Å, and 3.1 Å, respectively, as reported previously. 30 The average basal spacing for INH-Mg/Al LDH prepared by the ion exchange method (nanocomposite A) was found to be 12.00 Å. By subtracting the LDH layer thickness of 4.8 Å from the average basal spacing of 12.00 Å, we obtained 7.2 Å, which was exactly equal to the short (horizontal) axis of INH.…”

“…30 In brief, Mg(NO 3 ) 2 hexhydrated and Al(NO 3 ) 3 non-hydrated at a 1:2 ratio were dissolved in 250 mL of deionized water and stirred for 15 minutes while Mg/Al LDH was formed by raising the pH to 10 by the dropwise addition of a 2 mol/L sodium hydroxide aqueous solution. Next, 50 mL of a 1 mol/L solution of isoniazid was added to freshly prepared Mg/Al LDH and the sample was stirred for 2 hours.…”
