Comparison of Amounts and Properties of Collagen and Proteoglycans in Condylar, Costal and Nasal Cartilages

Abstract: In this study we compared the compositions of extracellular matrices of condylar, costal and nasal cartilages to characterize differences in growth patterns in relation to matrix composition. Condylar, costal and nasal cartilages of 25- and 35-day-old rabbits were extracted and subjected to bio- and histochemical analysis to determine total amounts of collagen and amounts and aggregating properties of proteoglycans. We found that proteoglycan content and aggregate formation were greatest in nasal cartilage, an… Show more

“…Total DNA content/sample was determined using a picogreen cell proliferation assay kit (Molecular Probes, Eugene, OR). Total GAG/sample was quantified using the Blyscan Glycosaminoglycan Assay kit (Pietila et al, 1999). Total collagen/sample was determined using a modified chloramine-T hydroxyproline assay (Woessner, 1961).…”

“…Total sGAG content was measured using the Blyscan Glycosaminoglycan Assay kit (Biocolor, Belfast, Northern Ireland), derived from 1,9-dimethyl-methylene blue binding. 33 Post hydrolyzation by 2 N NaOH for 20 min at 110°C, samples were assayed for total collagen content using a chloramine-T hydroxyproline assay. 42 The amount of type II collagen was quantified by a sandwich ELISA (Chondrex, Redmond, WA).…”

“…Total sulfated GAG was then quantified using the Blyscan Glycosaminoglycan Assay kit (Biocolor), based on 1,9-dimethylmethylene blue binding. 24,25 After being hydrolyzed by 2 N NaOH for 20 min at 1108C, samples were assayed for total collagen content by a chloramine-T hydroxyproline assay. 26 …”

“…31 Following preconditioning, specimens were exposed to incremental stress relaxation, whereby they were exposed to 15 min constant strain increments until failure. A maximum of 11 increments were allowed (4,8,12,16,20,25,35,50,70, 100, and 130% strain). A strain rate of 6 mm/min was employed between constant-strain increments.…”
