Abstract: Williams' medium E is superior to DMEM for transport studies in primary human hepatocytes. Supplementation with dexamethasone increase mRNA levels of NTCP and BSEP.

“…20,21 Since dexamethasone is a common additive to culture media, we have previously investigated their effect on bile acid transporters in primary hepatocytes and shown that dexamethasone addition in the cell culture media increased the expression of BSEP and NTCP. 22 In this study, we investigated how different substrates, collagen and matrigel, and the addition of dexamethasone to the culture media influence bile acid synthesis and secretion.…”

“…In addition, while BSEP inhibition in vesicles and hepatocellular transport for CLF give insights into the potential of a drug to affect the overall bile acid transport, the ability to separate compound effects specific to uptake (CL biliary ) and efflux (%BEI) is an important factor in assessing potential risk. However, toxic effects due to the build‐up of cytotoxic bile acids may not be adequately present in in vitro model systems (Mork et al, ) and other investigators have added bile acids to the culture media to circumvent this potential limitation (Chatterjee, Richert, Augustijns, & Annaert, ; Susukida, Sekine, Nozaki, Tokizono, & Ito, ). While the ability to observe these endpoints in a single model offers consistency that is not available across multiple test systems, a missing component from this model is an endpoint specifically related to mitochondrial dysfunction.…”
