Comparison of Diafiltration and Tangential Flow Filtration for Purification of Nanoparticle Suspensions

Abstract: The TFF, in concentration mode at TMP of 10 psi, is a relatively quick, efficient, and cost-effective technique for purification and concentration of a large nanoparticle batch (>or=200 ml). The DCD technique can be an alternative purification method for nanoparticle dispersions of small volumes.

“…Tangential flow filtration (TFF), or cross-flow filtration, has been found as a very practical method to purify nanoparticulate dispersions from these impurities (5,13,(17)(18)(19)(20). It is a more gentle and continuous process and more easily scaled up than, e.g., the traditional dead-end ultrafiltration (21) or ultracentrifugation (22).…”

“…Finally, MNT offer the possibility of long-term storage, favorably distinguishing MNT from those nanoparticle systems that require preparation a relatively short time prior to use. 53,54 Finally, because of the modular nature of MNT design, one could envisage utilizing this platform to generate personalized therapeutics by tailoring the ligand and drug profiles to the characteristics of individual patients.
