The effect of daidzein (D), 4-methylbenzylidene camphor (4-MBC) or estradiol-17b-benzoate (E 2 ) on muscle of osteoporotic rats during fracture healing was studied. After performing a metaphyseal tibia osteotomy in 96 osteoporotic 5-month-old female Sprague-Dawley rats, they received daily 50 mg D, 200 mg 4-MBC or 0 . 4 mg E 2 per kg body weight, or soy free (SF) diet up to 36 and 72 days. Mitochondrial activity, fiber area, and capillary density were analyzed in M. gastrocnemius. Osseous callus bridging of fracture was observed in half of the rats after 36 days. By day 72, fracture was healed in most of the animals. State 3 mitochondrial respiration significantly enhanced in E 2 , 4-MBC and D groups versus SF after 36 days (30, 32 and 32 vs 23 pmol O 2 /s per mg). It declined after 72 days, however, in E 2 group it was still at a higher level versus SF (25, 23 and 21 vs 20 pmol O 2 /s per mg). Size of fast oxidative glycolytic (FOG) and fast glycolytic (FG) fibers, capillary density did not differ significantly between the groups, however, at day 36 an increase in D and 4-MBC groups was detectable. FOG diameter was 64, 66, 68, and 58 mm and FG diameter was 88, 98, 95, and 89 mm in SF, D, 4-MBC, and E 2 groups. The ratio of capillaries to muscle fiber was 1 . 1, 1 . 4, 1 . 3, and 1 . 1 in SF, D, 4-MBC and E 2 groups by day 36. D and 4-MBC react similar to estrogen thereby improving oxidative cell metabolism in severe osteoporotic rats. The level of mitochondrial activity was higher, though no significant morphological differences could be shown.

Organic UV filters are frequently used in daily life, and hence are ubiquitously detected in the aquatic environment worldwide. Most monitoring efforts and toxicological studies are, however, limited to a few chemicals like benzophenone‐3 (BP‐3). In the present study, we chose other major organic UV filters, such as avobenzone (AVB), homosalate (HS), octisalate (OS), and octocrylene (OC), and reviewed information on their use, environmental occurrences, and endocrine disruption effects, available to date. Organic UV filters are used in high volume in many consumer applications, not only in sunscreen products but also in cosmetics, personal care products, home products, and food packaging. Environmental monitoring worldwide reveals that recreational coastal waters are among the hot spots of their contamination. An increasing number of experimental studies indicate that organic UV filters such as octinoxate (OMC), AVB, and HS may cause disruptions in sex hormones. Avobenzene and OMC can also influence thyroid function in experimental models. Observations in human population are rarely made, but OMC and OC have been associated with decreased androgenicity and increase of polycystic ovary syndrome, respectively. Further investigations are warranted to fill the knowledge gaps identified in the present study, to help develop relevant safety screening measures for organic UV filters. Ethylhexyl methoxycinnamate (EHMC) is a widely used UV filter present in a large number of personal care products (PCPs). Under normal conditions, EHMC occurs in a mixture of two isomers: trans-EHMC and cis-EHMC in a ratio of 99:1. When exposed to sunlight, the trans isomer is transformed to the less stable cis isomer and the efficiency of the UV filter is reduced. To date, the toxicological effects of the cis-EHMC isomer remain largely unknown. We developed a completely new method for preparing cis-EHMC. An EHMC technical mixture was irradiated using a UV lamp and 98% pure cis-EHMC was isolated from the irradiated solution using column chromatography. The genotoxic effects of the isolated cis-EHMC isomer and the nonirradiated trans-EHMC were subsequently measured using two bioassays (SOS chromotest and UmuC test). In the case of trans-EHMC, significant genotoxicity was observed using both bioassays at the highest concentrations (0.5 - 4 mg mL ). In the case of cis-EHMC, significant genotoxicity was only detected using the UmuC test at concentrations of 0.25 - 1 mg mL . Based on these results, the NOEC was calculated for both cis- and trans-EHMC, 0.038 and 0.064 mg mL , respectively. Risk assessment of dermal, oral and inhalation exposure to PCPs containing EHMC was carried out for a female population using probabilistic simulation and by using Quantitative in vitro to in vivo extrapolation (QIVIVE). The risk of cis-EHMC was found to be ∼1.7 times greater than trans-EHMC. In the case of cis-EHMC, a hazard index of 1 was exceeded in the 92nd percentile. Based on the observed differences between the isomers, EHMC application in PCPs requires detailed reassessment. Further exploration of the toxicological effects and properties of cis-EHMC is needed in order to correctly predict risks posed to humans and the environment. © 2016 Wiley Periodicals, Inc.
