Eggshell membrane (ESM), a plentiful biological waste, consists of collagen-like proteins and glycosaminoglycans (GAGs) such as hyaluronic acid (HA). Here we used a keratinase (oeMtaker)-mediated system to decompose ESM. The best reaction condition was established by incubating the solution containing oeMtaker, sodium sulfite, and ESM with a weight ratio of 1:120:600. ESM enzymatic hydrolysate (ESM-EH) showed a high proportion of essential amino acids and type X collagen peptides with 963–2259 Da molecular weights. The amounts of GAGs and sulfated GAGs in ESM-EH were quantified as 6.4% and 0.7%, respectively. The precipitated polysaccharides with an average molecular weight of 1300–1700 kDa showed an immunomodulatory activity by stimulating pro-inflammatory cytokines (IL-6 and TNF-α) production. In addition, a microorganism-based system was established to hydrolyze ESM by Meiothermus taiwanensis WR-220. The amounts of GAGs and sulfated GAGs in the system were quantified as 0.9% and 0.1%, respectively. Based on our pre-pilot tests, the system shows great promise in developing into a low-cost and high-performance process. These results indicate that the keratinase-mediated system could hydrolyze ESM more efficiently and produce more bioactive substances than ever for therapeutical applications and dietary supplements.

In this work, bioactive glass (BG) particles synthesized by a sol-gel method, hyaluronic acid (HYA) and collagen (COL) extracted from chicken eggshell membrane (ESM), and as-purchased polycaprolactone (PCL) were used to obtain a novel bioactive scaffold using the gel-pressing technique. Two composite mixtures in weight percent were obtained and identified as SCF-1 and SCF-2, and were characterized by using FTIR, XRD, and SEM techniques. Subsequently, the composite materials applied as coatings were evaluated in simulated body fluid solutions using electrochemical techniques. The results of bioactivity and biodegradability evaluations, carried out by immersing in simulated body fluid and phosphate-buffered saline solution, showed that the SCF-1 sample presented the best biocompatibility. In accordance with the potentiodynamic results, the 316L-SS and the SCF-1-coated SS showed a very similar corrosion potential (Ecorr), around −228 mV, and current density (icorr) values in close proximity, while the SCF-2-coated SS showed more positive Ecorr around −68 mV and lower icorr value in one order of magnitude. These results agree with those obtained by electrochemical impedance spectroscopy, which show a corrosion mechanism governed by activation and finite diffusion through the porous layer. In addition, results were complemented by dynamic compression testing under oscillating forces to identify the developed scaffolds’ response under external forces, where the SCF-1 scaffold presented a maximum compression. The degradation resistance, bioactivity, and mechanically obtained measurements provided interesting results for potential further studies in tissue engineering.

Hyaluronic acid from Liparis tessellatus eggs (HALTE) was grafted with caffeic acid (CA-g-HALTE), ferulic acid (FA-g-HALTE), gallic acid (GA-g-HALTE), and nisin (Nisin-g-HALTE) and investigated for their anti-inflammatory and antioxidant potential in lipopolysaccharides-stimulated RAW 264.7 mouse macrophages. Nitric oxide (NO) generation and prostaglandin E 2 activity were measured after treatment with the grafted HALTE samples. All grafted HALTE samples exhibited more antioxidant activity against 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radicals than 2,2-diphenyl-1-picrylhydrazyl radicals. Nisin-g-HALTE showed the least antioxidant activity. Additionally, the NO assay results showed that all grafted samples had no cytotoxic effect on RAW 264.7 macrophages and reduced macrophage activity after treatment. The most effective concentrations of CA-g-HALTE and FAg-HALTE were found to be above 100 μg/mL. Increased sample concentration resulted in increased activity except with Nisin-g-HALTE at 100 μg/mL. CA-g-HALTE, FA-g-HALTE, GA-g-HALTE, and Nisin-g-HALTE were found to have antioxidant and anti-inflammatory potential, which can be further explored for use in food, cosmetic, nutraceutical, and biomedical applications.
