Comparison of Ethanol Extract from Roots, Leaves, and Flowers of Calotropis gigantea as Anticancer on T47D Breast Cancer Cell Lines

Abstract: This research aims to find out the anticancer activity of ethanol extract from the roots, leaves, and flowers of Calotropis gigantea. This experiment was conducted by MTT method on T47D breast cancer cell line. The result showed that the root of Calotropis gigantea was more cytotoxic (IC50 89.76 μg/mL) on T47D breast cancer cell line than the leaves (IC50 459.51 μg/mL) and the flowers (IC50>1000). Based on the result, roots are potent to be chemotherapeutic agent, especially in breast cancer.

“…As reported by the researchers in a previous study, C. gigantea leaf ethanol extract can inhibit the growth of in vivo fibrosarcoma at doses of 100 and 150 mg/kg with the mechanism of increased expression of caspase-3 (Mutiah et al, 2016). Ethanol extract of the roots and C. gigantea leaves have an anticancer activity which is higher than the flower (Mutiah et al, 2016). Ethyl acetate fraction from the leaves (IC 50 41, 79 µg/mL) and dichloromethane fraction (IC 50 40.57 μg/mL) have cytotoxic activity which is higher than butanol fraction (IC 50 737.74 ug/mL) and water (IC 50 8493 ug/mL) (Mutiah et al, 2017).…”

“…After cell viability is known, a combination index is calculated. Combination index is a standard method used to evaluate drug combinations [14]. The smaller the value of a combination index, the more synergistic the combination will be -the results of the combination index calculation presented in Table 1.…”
