ABSTRAK Hasil: Periode konfluens rata-rata AT-MSCs pasca-radiasi adalah 4,33 hari. Tidak ada perbedaan bermakna viabilitas sel punca sebelum dan setelah paparan sinar-X (p=0,831). Tidak terdapat korelasi antara viabilitas sel punca sebelum kultur setelah dipaparkan berbagai dosis radiasi sinar-X (p=0,138, r=0,503). Tidak ada perbedaan bermakna waktu penggandaan populasi sel punca yang dikultur setelah paparan sinar-X dan sel punca tidak dikultur setelah paparan sinar-X dan di antara kelompok sel punca yang dipaparkan berbagai dosis sinar-X (p=0,792 ABSTRACTBackground: Adipose tissue derived mesenchymal stem cells (AT-MSCs) are relatively easy in isolation procedure compared to bone marrow-derived. : Human adipose derived stem cells (hADSCs) can be cultured in outdated platelet lysate (PL) containing medium. Processing of thrombocyte concentrate to get PL can be done by various freeze-thaw cycles. There was no information whether various processed PL containing media gave the same proliferation potential and surface marker expressions. Therefore, the aim of this study was to know the proliferation and surface marker characteristics of hADSCs after expansion in various processed PL containing media. hADCSs were cultured in various processed PL containing media, namely F1, F2, and F3, where the numbers denoted once, twice and three times freeze-thaw cycles. Proliferations were measured on day-2, day-4, day-7, and day-10. Positive surface markers were CD90, CD73, and CD105, while negative markers were a cocktail of CD34, CD45, CD11b, CD19, and HLA-DR. Difference in proliferations and surface marker expressions between F1, F2, and F3 were analyzed by one-way ANOVA. We found that cell proliferation in F1, F2, and F3 showed no significant difference on day-7 and day-10. The expression levels of CD90 and CD 105 in F1, F2, and F3 were all above 90%, which correspond with mesenchymal stem cells according to the requirement of International Society for Cell Therapy (ISCT). However, CD73 showed highest (97%) expression on F2 and lowest on F3 (81.8%). Negative marker range was 2.4-2.9%. In Conclusion, at the end of culture, hADSCs showed similar profile and proliferation, when cultured in F1, F2, and F3.

Flowcytometric analysis is widely used in characterization of stem cells. In areas where flowcytometry is not available, characterization of stem cells can be done using immunocytochemistry. This study aimed to compare the results of flowcytometry against immunocytochemistry for the measurement of CD34, CD73 and CD90 in stem cells. Adipose tissue derived stem cells were analyzed for their CD34, D73 and CD90 simultaneously, using a BD FACSCalibur flowcytometer (BD Biosciences) with 10,000 gated events. The same sample was made into spot specimens and checked for CD34, CD73 and CD90 by immunocytochemistry and when available, 100 cells were screened for positive results and percentages of positive results for CD34, CD73 and CD90 were computed. The results of the two methods were compared. Discrepancies between the two methods were noted for all samples. Discrepancies lower than 3, 11 and 11% for CD34, CD73 and CD90 respectively were regarded as agreement between the two methods. Percentages of agreement in CD34, CD73 and CD90 were calculated. Some spot specimens did not yield 100 cells and analyses were done on available cells. The agreement between the two methods for CD34, CD73 and CD90 were 22, 42 and 29% respectively. Agreement between flowcytometry and immunocytochemistry was low for CD34 and CD90 and was moderate for CD73.
