Comparison of Intraocular Treatment of DMTU and SOD Following Retinal Ischemia in Rats

Abstract: The effect of intravitreal injections of DMTU (dimethylthiourea) and SOD (superoxide dismutase), two free radical scavengers, was evaluated in a rat model of retinal ischemia induced by elevated intraocular pressure. The drugs were administered just before or just after a 60 min ischemia. At days 2 and 7 after reperfusion, retinal recovery was evaluated by electroretinography. At day 7, layer thicknesses and cell rows were measured from histologic sections of paraffin-embedded retinas. In the vehicle-treated c… Show more

“…Studies show that oxidative stress is a key player in retina neuronal injury in models of I/R [9–11]. To test whether WD-Hpt could reduce oxidative stress in I/R retina, we assessed formation of the peroxinitrite biomarker nitrotyrosine at 6 h after I/R by western blot analysis.…”

“…Oxidative stress is critically involved in neuronal cell death in ischemic retinopathy [9–11]. It causes diverse pathways such as inflammation [12, 13], proliferation [14] and apoptosis [15].…”

“…[31][32][33] NADPH oxidase is a major source of ROS in vascular cells. To understand the potential role of NADPH oxidase in neuronal injury in retinal I/R, the expression of different NOX isoforms and p47 phox and p22 phox subunits was analyzed at 6 hours after retinal ischemic injury.…”

“…Injury mediated by free radicals is a major cause of damage in ischaemic tissue after reperfusion (McCord, 1985), but it can be reduced by the use of antioxidants (Martz et al, 1989;Punch et al, 1991;Ambrosio & Flaherty, 1992;Karwatowska-Prokopczuk et al, 1992;Ernster, 1998;Rios et al, 1999). This is the first report of free oxygen radicals being generated at the time of oocyte aspiration from ovaries that have been stored for a long time (several hours), and these chemicals impair the developmental competence of oocytes.…”
