Comparison of ligand binding assay and enzyme immunoassay of oestrogen receptor in human breast cancer cytosols

Abstract: Nine laboratories cooperating in the Receptor Study Group of the EORTC compared results of ligand binding assay supplemented with Scatchard plot analysis (LBA) and enzyme immunoassay (EIA) for the assessment of oestrogen receptors in 1665 breast cancer cytosols. An excellent agreement was observed between results of Scatchard plot analysis and EIA in each laboratory. Linear correlation coefficients of log-transformed data (log [(ER + 10)/10]) ranged from 0.839 to 0.977 (n = 52-373; P less than 0.001); Spearman… Show more

“…The first part was submitted for histological study and histoprognostic grading according to the method of Contesso et al (8). The second part of the sample was immediately frozen in liquid nitrogen for estrogen receptor (ER) and progesterone receptor (PR) assays (2). The third part was used in molecular biologic assays (30).…”

“…Until 1989 the hormonal receptor status (estrogen and progesterone) was measured by ligand-binding assay [21]. Subsequently, measurements were obtained by cytosolic immunoassay using the Abbott kit [22]; these two methods had been previously compared and gave concordant results [23]. After 1997, hormonal receptor status was measured using the immunohistochemistry (IHC) method.…”

[35][36][37][38] Total RNA isolation Total RNA was extracted from cell cultures using the Quick RNA extraction kit (Talent SRL, Trieste, Italy) according to the manufacturer's specifications. Total RNA was isolated from tumor samples (40 mg weight) using the RNeasy Mini Kit (Qiagen, Courtaboeuf, France).…”
