Comparison of Ogawa Media, BACTEC MGIT 960 System and TB/NTM Real-Time PCR for DetectingMycobacteriumSpecies

Abstract: Background: Mycobacterial infection is a problem throughout the world along with the increase of immunocompromised patients. For this reason, there have been many methods for faster and more accurate diagnosis. In this study, we evaluated several laboratory methods for mycobacterial infection. In the Mycobacteria genotyping chip analysis, the most frequently identified NTM species in descending order were M. avium, M. intracellulare, M. chelonae and M. abscessus. Conclusion: Culturing with a combination of MGI… Show more

“…These conventional tests can take several weeks to perform and cannot always precisely identify the species6. Furthermore, these testing procedures are complex and laborious, and they are usually impeded by the slow growth of mycobacteria in culture.…”

“…The AdvanSure TB/NTM real-time PCR kit (LG Life Science, Seoul, Korea) was developed to detect MTB and non-tuberculous mycobacteria (NTM); it targets the insertion sequence, IS6110 and internal transcribed spacer (ITS) region, respectively. Although this kit showed high sensitivity and specificity [ 3 - 5 ], its competency for quantification of bacterial burden has not been studied. In this study, we evaluated the quantitative performance of AdvanSure TB/NTM assay by comparing cycle threshold (Ct) values with 1) acid-fast bacilli (AFB) staining grades and semi-quantitative colony counts on solid medium and 2) time to positivity (TTP) in solid and liquid media.…”
