Comparison of real-time PCR and conventional PCR for detection of Leishmania (Leishmania) infantum infection: a mini-review

Abstract: Abstract:In recent years, the polymerase chain reaction (PCR) technique has significantly advanced towards expanding its use and versatility by working with quantitative real-time PCR (qPCR). Data from the literature show that both methods present interesting characteristics for the diagnosis of visceral leishmaniasis. The benefits of qPCR in relation to conventional PCR include speed, reproducibility and quantitative ability. In addition to operational advantages, qPCR is more sensitive and reproducible and m… Show more

“…6,7 Of the four Leishmaniapositive samples, three had a previous negative PCR for Leishmania spp. performed before their inclusion in the study; this might be related to the lower sensitivity of the direct PCR method in comparison to RT-PCR 14,15,33,34 and/or to inappropriate selection of paraffin-embedded material or of the probes. Our results provide further support that RT-PCR, as previously documented, 14,15 is superior to immunohistochemistry in detecting Leishmania spp.…”

“…qPCR is considered to be a helpful tool for Leishmania diagnosis, monitoring during therapy, development of new drugs and diagnostic tools, comparison of drug efficacy or prophylactic schemes, and for epidemiological studies. Regarding diagnosis of leishmaniasis, the kinetic study of parasitemia in the immunocompromised hosts, the diagnosis of relapses and the quantification of the low parasitic load in asymptomatic patients are of great interest [47].…”
