Comparison of the Abbott RealTi m e CT New Formulation Assay with Two Other Commercial Assays for Detection of Wild-Type and New Variant Strains of Chlamydia trachomatis

Abstract: In an analytical-method comparison study of clinical samples, the Abbott RealTime CT new formulation assay (m2000 real-time PCR), consisting of a duplex PCR targeting different parts of the cryptic plasmid in Chlamydia trachomatis, was compared both with version 2 of the Roche Cobas TaqMan CT assay, comprising a duplex PCR for a target in the cryptic plasmid and the omp1 gene, and with the Gen-Probe Aptima Combo 2 assay (AC2) targeting the C. trachomatis 23S rRNA molecule. First-catch urine samples from Sweden… Show more

“…In order to ensure accuracy of NAAT results, strategies such as utilization of dual targets in the assays (16,17) or inclusion of a supplemental "confirmation" have been incorporated in the testing protocol (15,18,19). These approaches are, in particular, important for detection from the rectum and oropharynx, which is crucial particularly in population groups of men who have sex with men (20) or with history of oral and/or anal sex.…”

“…in the SF of patients with CIA and related diseases (reviewed in [21]). In previous studies we have shown that the sensitivity of PCR and ligase chain reaction (LCR) basically depends on sample preparation, as well as the amplification process itself [7,12,14]. However, detection of C. tr.…”

“…Earlier studies using commercially available amplification tests and in-house PCR in UA patients identified Chlamydia in only 2-25% of the analyzed SF samples [8,15,18]. Commercial assays have improved during past years, and now display very high sensitivity and use very efficient preanalytical steps [12]; nevertheless none of these tests has been tested specifically in SF and approved for diagnosing CIA.…”
