Comparison of the aneugenic effect of vinorelbine and vincristine in cultured human lymphocytes

Abstract: Vinca alkaloids are used clinically against a variety of hematological and solid tumors. These compounds interact with tubulin subunits to prevent microtubule assembly, inducing abnormal chromosome segregation in dividing cells and causing aneuploidy. The vinca alkaloid vincristine sulfate (VCR) and the semisynthetic analog vinorelbine (VRB) were studied by analysis of micronuclei (MN) in cultured human lymphocytes using the cytokinesis block method. Furthermore, fluorescence in situ hybridization with a human… Show more

“…Sterile water was used as the negative control. The mutagenic compounds were used at the following final concentrations: EMS (Sigma), 120 mg/ml [Fimognari et al, 2001]; VIN (Sigma), 0.05 mg/ml [González-Cid et al, 1999]; H 2 O 2 (Sigma), 3.4 mg/ml [Fenech et al, 1999a]; MMC (Sigma), 0.1 mg/ml [Caria et al, 1997].…”

“…The induction of MnCþ could be attributed to the ability of Al to modify the structure of tubulin (Sivaguru et al, 2003). as it is well known that agents that damage mitotic spindles induce aneuploidy (Gonzalez-Cid et al, 1999).…”

“…As different investigators relate MN appearance with polyploidy and aneuploidy in mammalian cells caused by different genotoxic compounds or treatment (Ciranni et al, 1995;Uryvaeva and Delone, 1995;Elhajouji et al, 1998;Miller et al, 1998;Gonzalez-Cid et al, 1999), the aim of our study was to establish MN appearance in triploid rainbow trout. To the best of the author's knowledge this is the first study of this kind conducted on triploid fish.…”
