Comparison of the Efficacy and Safety of Pantoprazole Magnesium and Pantoprazole Sodium in the Treatment of Gastro-Oesophageal Reflux Disease

Abstract: Pantoprazole-Mg is clinically as effective and well tolerated as pantoprazole-Na in the treatment of GORD stages I-III, demonstrating non-inferiority for oesophageal healing at 8 weeks and superior healing rates at 4 weeks associated with high levels of symptomatic relief.

“…Although proton pump inhibitors (PPIs), the most potent therapeutic option for GERD, have been shown to improve night-time symptoms of GERD, some patients continue to experience breakthrough symptoms 4. This may reflect a wearing off of the acid suppressive effect of PPIs towards the end of the dosing interval, due to the rebound of acidity levels through a combination of proton pump regeneration and the short half-life of PPIs 5,6…”

“…At 8 weeks, healing rates with pantoprazole‐Mg were similar to those with pantoprazole‐Na (87.3% vs. 85.0%); however, after 4 weeks of treatment, pantoprazole‐Mg achieved a superior healing rate over that with pantoprazole‐Na (72.7% vs. 66.2%). This was associated with numerically higher rates of symptom relief across a broad range of symptoms at 4 weeks in the pantoprazole‐Mg group than those in the pantoprazole‐Na group …”

“…Pantoprazole‐Mg has been shown to be at least as effective as pantoprazole‐Na in oesophageal healing and relief of a broad range of gastrointestinal symptoms associated with GERD . At 8 weeks, healing rates with pantoprazole‐Mg were similar to those with pantoprazole‐Na (87.3% vs. 85.0%); however, after 4 weeks of treatment, pantoprazole‐Mg achieved a superior healing rate over that with pantoprazole‐Na (72.7% vs. 66.2%).…”
