Comparison of two sampling techniques to assess quantity and distribution ofMalasseziayeasts on the skin of Basset Hounds

Abstract: Cytological examination using the tape-strip technique and fungal culture using contact plates with modified Dixon's medium were compared to evaluate the carriage of Malassezia yeasts on four cutaneous sites (left pinna, umbilical region, axilla and perianal area) in adult Basset Hounds. Twenty animals were included in the study. High numbers of Malassezia were isolated from at least one area in 100% of the animals. The frequencies of isolation and population sizes differed significantly according to anatomica… Show more

“…31 Various methods have been used to collect material for cytological and cultural examinations of Malassezia species. Cytological techniques include impression methods using glass slides, cotton swabs, skin scraping, and tape strip preparation, 1,23,32 whereas mycological cultures may be obtained from cotton swabs or directly with contact plates. 2 Some of these techniques may be less sensitive than others, but none has yet been accepted as a standard.…”

“…2 Some of these techniques may be less sensitive than others, but none has yet been accepted as a standard. 1,2,6,23,32 On the basis of the empirical evaluation of this study, cotton swabs were considered to be the most reliable technique because they may be used for both cytological examination and culture and are easy to use. It is well known that cytological examinations of the ear or skin of dogs detect a small number of commensal organisms.…”

“…Lesions affecting the front feet, ear pinnae, ear pinna margins and dorsolumbar area were noted. After physical examination, the skin was sampled for the presence of Malassezia, by the acetate tape method applied to both axillae, both inguinal areas, and a skin lesion, if present (Bensignor et al, 2002;Machado et al, 2011).…”

“…Sample collection and processing-For each dog, clear cellophane adhesive tape was used as described 6,7 to obtain a sample from the skin surface of the upper and lower eyelids of each eye for cytologic evaluation. Briefly, the upper and lower eyelids were held closed and a strip of adhesive tape was placed against the periocular skin of both eyelids simultaneously; gentle pressure was applied to the tape to ensure contact with the skin.…”
