Schisandrae Sphenantherae Fructus (SSF), the dry ripe fruit of Schisandra sphenanthera Rehd. et Wils., is a traditional Chinese medicine with wide application potential. The quality of SSF indicated by the composition and contents of secondary metabolites is closely related to environmental factors, such as regional climate and soil conditions. The aims of this study were to predict the distribution patterns of potentially suitable areas for S. sphenanthera in China and pinpoint the major environmental factors influencing its accumulation of medicinal components. An optimized maximum entropy model was developed and applied under current and future climate scenarios (SSP1-RCP2.6, SSP3-RCP7, and SSP5-RCP8.5). Results show that the total suitable areas for S. sphenanthera (179.58×104 km2) cover 18.71% of China’s territory under the current climatic conditions (1981–2010). Poorly, moderately, and highly suitable areas are 119.00×104 km2, 49.61×104 km2, and 10.98×104 km2, respectively. The potentially suitable areas for S. sphenanthera are predicted to shrink and shift westward under the future climatic conditions (2041–2070 and 2071–2100). The areas of low climate impact are located in southern Shaanxi, northwestern Guizhou, southeastern Chongqing, and western Hubei Provinces (or Municipality), which exhibit stable and high suitability under different climate scenarios. The contents of volatile oils, lignans, and polysaccharides in SSF are correlated with various environmental factors. The accumulation of major secondary metabolites is primarily influenced by temperature variation, seasonal precipitation, and annual precipitation. This study depicts the potential distribution of S. sphenanthera in China and its spatial change in the future. Our findings decipher the influence of habitat environment on the geographical distribution and medicinal quality of S. sphenanthera, which could have great implications for natural resource conservation and artificial cultivation.

Schisandra sphenanthera Rehd. et Wils., as a traditional Chinese medicine, has important medicinal value. In the market, the availability of the fruit of S. sphenanthera mainly relies on wild picking, but many canes and leaves are discarded during wild collection, resulting in a waste of resources. The canes and leaves of S. sphenanthera contain various bioactive ingredients and can be used as spice, tea, and medicine and so present great utilization opportunities. Therefore, it is helpful to explore the effective components and biological activities of the canes and leaves to utilize S. sphenanthera fully. In this study, the response surface method with ultrasound was used to extract the total triterpenoids from the canes and leaves of S. sphenanthera at different stages. The content of total triterpenoids in the leaves at different stages was higher than that in the canes. The total triterpenoids in the canes and leaves had strong antioxidant and antibacterial abilities. At the same time, the antibacterial activity of the total triterpenoids against Bacillus subtilis and Pseudomonas aeruginosa was stronger than that against Staphylococcus aureus and Escherichia coli. This study provides the foundation for the development and utilization of the canes and leaves that would relieve the shortage of fruit resources of S. sphenanthera.

Schisandra sphenanthera is an very important medicinal plant, and its main medicinal component is bioactive lignans, which has been developed over the years as an important cash crop in the central mountainous. The S. sphenanthera fruit has the characteristics of food and medicine homology, which is favored by the majority consumers, but the root, stem, and leaf are not fully used. Furthermore, the genetic characteristics of S. sphenanthera are rarely understood, thus hindering the study of functional genome and lignan biosynthesis. To better understand the lignan metabolic pathway, transcriptome and metabolome analysises were performed on four major tissues in S. sphenanthera. As a consequence, 167,972,229 transcripts and 91,215,760 unigenes with an average length of 752 bp were identified. Tissue-specific gene analysis revealed that the abundance of unique unigenes was highest in roots (9, 703), and lowest in leaves (189). Transcription factor analysis showed that MYB-, bHLH- and ERF-transcription factors, which played important roles in the regulation of secondary metabolism, showed rich expression patterns and may be involved in the regulation of the lignan metabolic processes. In the different tissues, lignans were preferentially enriched in fruits and roots by the genes expression profiles related to lignan metabolism and lignan compounds relative content. Furthermore, schisandrin B was found to be an important compound in S. sphenanthera. According to WGCNA analysis, PAL1, C4H-2, CAD1, CYB8, OMT27, OMT57, MYB8, bHLH3, and bHLH5 could be related to the accumulation of lignans in S. sphenanthera fruits, CCR5, SDH4, CYP8, CYP20 and ERF7 could be related to the accumulation of lignans in S. sphenanthera roots. In this study, the transcriptome sequencing and targeted metabolic analysis of lignins that will lay a foundation for further study of biosynthetic genes of lignin and other natural products in S. sphenanthera, and also provided a new idea for the rational utilization of different tissues of S. sphenanthera.
