Competence for genetic transformation in encapsulated strains of Streptococcus pneumoniae: two allelic variants of the peptide pheromone

Abstract: The nucleotide sequence of comC, the gene encoding the 17-residue competence-stimulating peptide (CSP) of Streptococcus pneumoniae (L. S. Håvarstein, G. Coomaraswamy, and D. A. Morrison, Proc. Natl. Acad. Sci. USA 92:11140-11144, 1995) was determined with 42 encapsulated strains of different serotypes. A new allele, comC2, was found in 13 strains, including the type 3 Avery strain, A66, while all others carried a gene (now termed comC1) identical to that originally described for strain Rx1. The predicted matur… Show more

“…The resulting plasmid pJY4164-ply was transformed into S. pneumoniae strain WU2 by using competencestimulating peptide 2 (as described in ref. 35; synthesized by the Biopolymer Facility at Harvard University) at a concentration of 100 ng͞ml by using protocols as described (36). Colonies that were resistant to erythromycin (0.3 g͞ml) were screened for pneumolysin production by measuring the hemolytic activity of supernatants from overnight cultures in Todd-Hewitt medium supplemented with 0.5% yeast extract.…”

“…The capsular variants of otherwise isogenic strains were constructed in three different genetic backgrounds-TIGR4 (originally of serotype 4) and 603 and 618 (both originally of serotype 6B)-by using a previously described method (48). In short, the parent strain was rendered unencapsulated by insertion of a cassette into the capsule locus using the transformation protocol published by Pozzi et al (37) and the bicistronic Janus cassette constructed by Sung et al (46). Encapsulated mutants were generated by replacing the cassettes with a cps locus from a donor strain.…”

“…Complementation of mutants VES2585-VES2587 by pDLP49, carrying an intact copy of the tts gene under the control of its own promoter (Llull et al, 2001), was performed by a similar transformation method, but adding competence-stimulating peptide 2 (CSP-2) (Pozzi et al, 1996) to the transformation mix at a final concentration of 100 ng ml 21 . CSP-2 was used instead of CSP-1 after empirical confirmation of its ability to induce competence in type 37 derivatives.…”
