Complement C3 and C4, but not their regulators or activated products, are associated with incident metabolic syndrome: the CODAM study

Abstract: PurposeWe investigated the associations of components of the alternative (C3, C3a, Bb, factor D [FD], factor H [FH], properdin) and the classical complement pathway (C4, C1q, C1-inhibitor [C1-INH]) with prevalent and incident metabolic syndrome in a cohort with a moderately increased risk of cardiometabolic disease.MethodsThe study cohort was comprised of 574 participants (61% men, age 59.6 ± 7.0 years) at baseline and 489 participants after 7-year follow-up. Multiple logistic regression analyses were done to … Show more

“…A study conducted among people of Caucasian descent above 40 years showed significant associations of C1q with TG, HDL-C, and FPG. As reported in that study, the relationship between C1q and MetS was modest (42), which was inconsistent with our results. This difference may be attributed to the subjects belonging to a specific population of individuals who already exhibit moderately increased risk of cardiometabolic diseases.…”

Complement system is considered that defense system for resistance against microbial infection, mainly included C3 and C4 [36] . “…Some studies also showed a significant improvement in homeostasis assessment for the insulin resistance index after lowering the level of SUA (Liu et al, 2015;Takir et al, 2015). Previous researches revealed that the levels of serum C3 and C4 positively correlated with the development of the metabolic syndrome and had been identified as important markers relevant to this disease (Meng et al, 2017;Meng et al, 2018;Xin et al, 2018). The increased activation products of C3 can accelerate the uptake of free fatty acids, the synthesis of triacylglycerol, and inhibit hormone-sensitive lipase in several adipocytes, thus contribute to the development of the metabolic syndrome (Phieler et al, 2013;Barbu et al, 2015).…”
