Complete mitochondrial genome of the freshwater gudgeon,Microphysogobio koreensis(Cypriniformes, Cyprinidae)

Abstract: The complete mitochondrial genome was sequenced from the freshwater cyprinid fish, Microphysogobio koreensis. The genome sequence was 16,606 bp in size, and the gene order and contents were identical with other cyprinid species. Of 13 protein-coding genes (PGCs), three genes (CO2, CO3, Cytb) had an incomplete stop codon as shown in other cyprinid fishes. The base composition of M. koreensis showed anti-G bias (13.79% and 10.16%) on the second and third positions of PCGs.

“…Their average base composition was as follows: 34.2% A, 20.0% T, 21.2% G, and 24.6% C. The average A + T content of both rRNAs was 54.2% (Table 2). The lengths and A + T content of these two rRNAs were well within the ranges observed in other Microphysogobio mitogenomes (Lin et al 2014;Hwang et al 2014;Cheng et al 2015).…”

“…All mitochondrial genes were encoded on the heavy strand (H strand), except the ND6 gene and eight tRNAs (Table 1). The arrangement and content of these genes were conserved and typical of Microphysogobio mitochondrial genomes (Hwang et al 2014;Lin et al 2014;Cheng et al 2015). The M. elongatus mitogenome contained a total of 21 bp overlapping regions which were in six pairs of neighboring genes, ranging from 1 to 7 bp in length.…”

“…It contained 13 protein-coding genes, two ribosomal RNA genes, 22 transfer RNA genes, one displacement loop (D-loop) locus and an origin of replication on the light-strand (OL). In these genes, ND6 and eight tRNA genes were encoded on light strand while the rest of genes were encoded on heavy strand and the arrangement of these genes were the same as that found in teleosts (Hwang et al 2014;Cheng et al 2015). The overall base composition was 30.78% A, 26.09% T, 26.59% C, and 16.54% G, with a slight AT bias (56.87%).…”

“…Five PCGs (CO1, ATPase 8, ND4L, ND5, and ND6) were stopped with a complete 'TAA,' four PCGs (ND2, ATPase 6, ND3, and ND4) were stopped with an incomplete 'TA-.' Three PCGs (CO2, CO3, and Cytb) have an incomplete 'T-,' a common feature indicating the termination codon in Microphysogobio (Hwang et al 2014;Park et al 2016;Wang et al 2016). The overall base composition of the M. rapidus genome was 29.96% A, 26.06% T, 17.24% G, and 26.74% C, with high A þ T content of 56.02%.…”
