Complete nucleotide sequence of alfalfa mosaic virus RNA 2

Abstract: Double-stranded cDNA of in vitro polyadenylated alfalfa mosaic virus (AlMV) RNA 2 has been cloned and sequenced. The use of an oligodeoxyribonucleotide corresponding to the known sequence of the 5'-end of RNA 2 to prime second-strand DNA synthesis, enabled us to construct the complete primary structure of AlMV RNA 2. The sequence of 2,593 nucleotides contains a long open reading frame for a protein of Mr 89,753 starting at the first AUG codon from the 5'-end. This coding region is flanked by a 5'-terminal sequ… Show more

“…The mass of protein synthesized was calculated from the amount of[35S]methionine in the bands and the number of methionine residues calculated to be in each protein (i.e. 18, 16, 7 and 2 respectively for PI, P2, P3 and P4) from the sequences of RNA 1 (strain 425, Cornelissen et al, 1983a), RNA 2 (strain 425, Cornelissen et al, 1983b) and RNA 3 (S-strain, Ravelonandro et al, 1984), allowing for the removal of the N-terminal methionine residue from coat protein P4 synthesized in the wheat germ extracts (Thivent, 1983).…”

“…The RNA-2 of members of the Bromoviridae encodes a polymerase (Ahlquist et al, 1984;Cornelissen et al, 1983;Rezaian et al, 1984). As RNA dependent polymerases are probably among the most ancient of viral enzymes and appear to be more highly conserved than coat proteins, they are potentially 'the best ruler' for molecular taxonomy of RNA viruses (Bruenn, 1991).…”

“…Tobacco plants (Nicotiana tabacum L.var. Samsun NN) were inoculated with alfalfa mosaic virus, a plant virus with a tripartite genome (3) consisting in total of 8274 bases (2,5,6). We used our standard strain AMV 425 which is fully adapted to tobacco.…”
