Complete Sequence, Analysis and Organization of the Orgyia leucostigma Nucleopolyhedrovirus Genome

Abstract: The complete genome of the Orgyia leucostigma nucleopolyhedrovirus (OrleNPV) isolated from the whitemarked tussock moth (Orgyia leucostigma, Lymantridae: Lepidoptera) was sequenced, analyzed, and compared to other baculovirus genomes. The size of the OrleNPV genome was 156,179 base pairs (bp) and had a G+C content of 39%. The genome encoded 135 putative open reading frames (ORFs), which occupied 79% of the entire genome sequence. Three inhibitor of apoptosis (ORFs 16, 43 and 63), and five baculovirus repeated … Show more

“…Most baculovirus genomes sequenced to date contain from 1 to 16 hrs [50]. These DNA elements are interspersed throughout the genome and have been implicated as putative origins of DNA replication ( oris) , as enhancers of gene transcription, and in homologous recombination [51].…”

“…Five representative baculoviruses were chosen for the comparative study of SujuNPV: Autographa californica MNPV (AcMNPV, group I alphabaculovirus) [25] , Helicoverpa armigera SNPV (HearNPV, group II alphabaculovirus) [26] , Cydia pomonella GV (CpGV, betabaculovirus), Neodiprion lecontei NPV (NeleNPV, gammabaculovirus) and Culex nigripalpus NPV (CuniNPV, deltabaculovirus). SujuNPV shared 102 ORFs with AcMNPV, 108 with HearNPV, 78 with CpGV, 43 with NeleNPV, and 39 with CuniNPV, with an average amino acid (aa) identity of 36.0%, 39.0%, 28.4%, 23.0% and 16.3%, respectively.…”
