Complex Diversity in Ligularia Kanaitzensis

Abstract: Two samples of Ligularia kanaitzensis collected in the Shizhongshan Mountain region of Yunnan Province, as well as one sample collected in Sichuan Province, were analyzed with regard to root constituents and neutral DNA sequences. The two Shizhongshan samples were found to be different from each other and from other samples analyzed previously, indicating that the intra-specific diversity in the species was highly complex.

“…The sequence of sample 3 was consistent with the sample being a hybrid of L. vellerea and L. kanaitzensis (sample 6). From the atpB-rbcL sequence of sample 3, its maternal parent was judged to be L. vellerea, as the sequences were distinct between L. vellerea (sample 1, see also [4]) and L. kanaitzensis (sample 6, see also [7,8]). The parental lineage of sample 2 could not be deduced, as the atpB-rbcL sequence was similar to that in L. cyathiceps [9] and L. vellerea [4] (Table 2).…”

“…The 2 methyl groups of the senecioate (= 3-methylbut-2-enoate) moiety in 6-senecioyloxy-1,10-epoxyfuranoeremophilane was observed at δ 2.13 ( Z -Me) and 1.43 ( E -Me) in C 6 D 6 . 17 HMBC correlation was observed between H-6 and C-1″ for both 1c and 1d , supporting that 1a , 1c , and 1d are variants of the acyloxy group at C-6.…”

“…In the hope of obtaining a clearer picture of the phylogenetic structure of L. kanaitzensis , we carried out analysis on the present and the previous samples, [9,10,27] totaling 27, listed in Table S3 . For this purpose, we re‐analyzed the samples whose sequences remained ambiguous using a new set of primers [9,10,28] . The ITS1‐5.8S‐ITS2 sequences are summarized in Tables S4 .…”
