Complex Quantitative Traits Cracked by the Mouse Inter-Subspecific Consomic Strains

Abstract: Mammalian quantitative traits that are observed at the whole-body level, such as body weight and length and blood biochemical parameters, are determined by the cooperative effects of multiple genetic and epigenetic factors as well as environmental factors. This complexity has hampered the genetic analysis of quantitative traits. To overcome this difficulty, we have established a full set of consomic mouse strains, also known as chromosome substitution strains, by replacing every chromosome of the classical inb… Show more

“…S1). A large number of intersubspecific SNPs and indels detected in this study underlie the large phenotypic differences between M. m. domesticus and M. m. molossinus (Takada et al 2008;Takahashi et al 2008;Koide et al 2011;Takada and Shiroishi 2012). In addition, we identified a much larger number of nonsynonymous SNPs leading to radical amino acid substitutions (GMS > 150) than previously detected between the FVB/NJ and B6 laboratory strains (Wong et al 2012), and this is likely due to the intersubspecific genome divergence between M. m. molossinus and M. m. domesticus, from which the genomes of the classical laboratory strains are predominantly derived.…”

“…A panel of B6-ChrNMSM consomic strains were established and has been maintained at NIG. The process used to establish this panel was described previously (Takada et al, 2008; Takada and Shiroishi, 2012); briefly, MSM was backcrossed into B6 over 10 generations, and all consomic strains have the same genetic background as B6 except for one pair of chromosomes, which have been substituted for the corresponding MSM chromosome.…”

“…In other words, a large number of interesting genes are still yet to be identified in MSM/Ms. Genetic resources are already established with MSM/Ms, such as the complete genome sequence information [21], a full set of mouse consomic strains [61], the BAC clone library [24], Microsatellite database [23], and ES cells [37]. Using currently available genetic tools developed from MSM/Ms will allow us to identify responsible genes and polymorphisms for complex traits including cancer, which were previously difficult to handle.…”
