Comprehensive analysis of pan‐cancer reveals potential of ASF1B as a prognostic and immunological biomarker

Abstract: Background Anti‐silencing function 1 (ASF1) is a conserved histone H3–H4 chaperone protein. ASF1B, a paralog of ASF1, acts by promoting cell proliferation and influencing cell cycle progression. Although there is some evidence demonstrating that ASF1B plays a key role in the development, progression, and prognosis of certain cancers, there are no pan‐cancer analyses of ASF1B. Methods We used a range of bioinformatics approaches to investigate the predictive role of ASF1B, including its correlation with prognos… Show more

“…Recently, an increasing number of studies have found a correlation between cancer progression and tumor immune infiltration ( 30 , 31 ). Through public data mining, we also found a certain correlation between the expression of ASF1B and the immune infiltration of HCC, which is consistent with the conclusions of current reports ( 28 , 29 ). But the specific experiments need to be further verified.…”

“…In this research, we used bioinformatics and multiple databases to comprehensively analyze the expression of ASF1B in pan-cancer, and found that ASF1B was abnormally expressed in most cancers, including liver cancer, which is consistent with current literature reports (27)(28)(29). In order to explore the relationship between ASF1B and HCC in detail, RNA-seq data and corresponding clinical data of HCC in TCGA were analyzed separately.…”

“…In addition, several studies have underlined the important prognostic impact of ASF1B on a variety of cancers ( Corpet et al, 2011 ; Chen Z. et al, 2021 ; Feng et al, 2021 ). Another exciting finding was that ASF1B could regulate immune infiltration through affecting immune-related genes and pathways such as antigen processing and presentation and natural killer cell-mediated cytotoxicity ( Hu et al, 2021 ; Zhan et al, 2021 ), conferring its strong hints on potential immunotherapeutic target for several malignancies. The both overall and disease free survival analysis also demonstrated that the high expression of ASF1B was associated with high risk ( Figures 4C , 5C ), suggesting it serving as a tumor promoter in pancreatic adenocarcinoma.…”

“…Immunotherapy for tumors has recently emerged as a new approach to oncology treatment, specifically immune checkpoint blockade (ICB) therapy [ 4 ]. The emergence and refinement of gene expression databases have enabled explorations of new immunotherapeutic targets through pan-cancer analysis of the expression of particular gene and assessments of its relevance to patients' prognoses and potential mechanisms [ 5 , 6 ].…”

“…The infiltration levels of TIIC in TME also impact on the prognoses of cancers. For example, high levels of B, CD4+, and dendritic cells infiltration are associated with a better outcome in thymomas, which may be partially regulated by ASF1B [ 6 ]. Lower grade glioma (LGG) patients with high TUBA1C expression may have a better response to ICB [ 18 ].…”
