Comprehensive analysis of PTPN family expression and prognosis in acute myeloid leukemia

Abstract: Background: Tyrosyl phosphorylation is carried out by a group of enzymes known as non-receptor protein tyrosine phosphatases (PTPNs). In the current investigation, it is hoped to shed light on the relationships between the expression patterns of PTPN family members and the prognosis of acute myeloid leukemia (AML). Methods:PTPN expression was examined using GEPIA and GEO databases. To investigate the connection between PTPN expression and survival in AML patients, we downloaded data from the Broad TCGA Firehos… Show more

“…PTPN20 has been shown to have differential expression across cancer types, suggesting that it may have distinct roles in different cancer types. Specifically, while the expression of PTPN20 was reduced in colorectal cancer (19), PTPN20 levels showed no change between acute myeloid leukemia cell and normal cells (41). In esophageal carcinoma, PTPN20 was upregulated and associated with both the TNM stage and N stage.…”

“…According to the ProteinAtlas database, neutrophil cytosol factor 1 (NCF1), integrin beta-2 (ITGB2/CD18), tyrosine-protein phosphatase nonreceptor type 6 (PTPN6), and WAS/WASL-interacting protein family member 1 (WIPF1) are mainly expressed in lymphoid tissue, bone marrow, spleen, and myeloid blood cells. On the other hand, a high expression of PTPN6 and SAMHD1 [27,28] correlates with poor prognosis for AML patients. These molecules could play a role in the mechanism of resistance to retinoid-based therapy.…”

“…Concurrently, Human embryonic kidney 293 cells (Lenti-X-293T) (ATCC) were sustained in Dulbecco’s Modified Eagle’s Medium (Invitrogen), supplemented with 10% FBS and 100 IU/mL penicillin/streptomycin. Culturing conditions were set to 37°C with a humidified atmosphere consisting of 95% air and 5% CO 2 , following previously established protocols [ 7 ]. Periodic checks were conducted to ensure the cell cultures were devoid of mycoplasma contamination.…”
