Comprehensive Analysis of REST/NRSF Gene in Glioma and Its ceRNA Network Identification

Abstract: We sought to clarify the clinical relationship between REST/NRSF expression and the prognosis of glioma and explore the REST-associated competitive endogenous RNA (ceRNA) network in glioma. We downloaded RNA-seq, miRNA-seq and correlated clinical data of 670 glioma patients from The Cancer Genome Atlas and analyzed the correlation between REST expression, clinical characteristics and prognosis. Differentially expressed genes (DEGs) were identified with DESeq2 and analyzed with Gene Ontology (GO) and the Kyoto … Show more

“…1A. Consistent with other published analyses [6], higher REST expression was associated with worse overall survival in the pooled population of patients with low-grade glioma (LGG) and glioblastoma (logrank p = 9.9e − 11, Fig. 1B).…”

“…One of the deregulated genes in GBM is a repressor element-1 silencing transcription factor (REST), a transcriptional repressor that has been identified as an oncogenic protein in various brain tumor types, including neuroblastoma, medulloblastoma, and glioblastoma [4,5]. High expression of REST was significantly associated with worse overall survival, progression-free interval, and worse disease-specific survival in glioma patients [6]. Targeting REST may inhibit cancer stem cell proliferation as REST is crucial for cancer stem cell self-renewal [7].…”

“…Quantitative PCR was performed using QuantStudioTM 5 Real-Time PCR System with Hieff ® qPCR SYBR Green Master Mix (YEASEN Biotech Co., Ltd., China). The real-time qPCR was performed as previously described ( Zhang et al, 2020a ; Zhang et al, 2021d ). The corresponding mRNA levels were normalized to GAPDH as an internal control by the 2 −ΔΔCt method.…”

“…One study revealed that REST may downregulate the expression of SYN1 in glioma, thus maintaining a cancer stem-like phenotype and contributing to the development of glioma 18 . Zhang et al 19 found that higher REST level was significantly associated with worse clinical characteristics and poor clinical outcomes in patients suffering from glioma. A REST-associated competitive endogenous RNA (ceRNA) network including NR2F2-AS1-miR129-REST and HOTAIRM1-miR137-REST was constructed based on the TCGA and Starbase 19 .…”
