Concentration of anti-pneumococcal capsular polysaccharide IgM, IgG and IgA specific antibodies in adult blood donors

Abstract: ObjectivesAnti-pneumococcal capsular polysaccharide (PCP) IgM, IgG and IgA ELISAs have been developed to aid assessment of the adaptive immune system. The relationship between the concentrations of PCP IgM, IgG, and IgA was investigated.Design and methodsThe concentrations of PCP IgM, IgG, and IgA were measured in sera obtained from 231 adult blood donors.ResultsConcentrations of each isotype were not normally distributed. The median concentration for PCP IgM was 54 U/mL (range 37–75 U/mL), IgG 40 mg/L (range … Show more

“…[3][4][5] The IgA and IgM responses to pneumococcal vaccination have been reported to be compromised in some pediatric patients with antibody deficiencies [6] and correlates with common variable immunodeficiency (CVID) patients and their history of infection. [7] Using reported unknown vaccination reference ranges [8] we measured the concentrations of Pneumococcal IgA and IgM responses to Pneumovax in individuals diagnosed with antibody deficiencies who had normal and abnormal pneumococcal capsular polysaccharide (PCP) IgG concentrations.…”

“…Twelve patients (60%) had significant IgG antibody concentration, 15 patients (75%) had IgG2, 12 patients (60%) had IgM, and 5 patients (25%) had IgA. The threshold antibody level correlated with a protection from invasive disease is available in Rose et al for IgG and IgG2 and Parker et al for IgA and IgM: 51.5 µg/mL IgG, 19.5 µg/mL IgG2, l50 UI/mL IgA, and 20 U/mL IgM, respectively.…”

“…[14] Adult blood donor samples (n = 79, median age 55 years, range 18-90 years) were collected in donor centres by Biomex Solutions (Heidelberg, Germany) and purchased from Quest Biomedical (Solihull, UK). [15] (2) Control Group B consisted of matched pre and post-vaccination samples from healthy adults vaccinated with Pneumovax (n = 10, 20 samples [16] ). For control group B, post vaccination samples were taken 28 days post vaccination (16) and for patient groups A-C samples were taken during routine immunological investigation 3-8 weeks post vaccination.…”
