Confirmation of 11-nor-Δ9-Tetrahydrocannabinol in Urine as Its t-Butyldimethylsilyl Derivative Using GC/MS

Abstract: A procedure for the detection and quantitation of 11-nor-delta 9-tetrahydrocannabinol-9-carboxylic acid, the major metabolite of delta 9-tetrahydrocannabinol, in urine is presented. Because a significant portion of the metabolite is present as a conjugated form, the urine was hydrolyzed by the addition of strong base. The solution was then acidified and the metabolite extracted into an organic solvent. It was subsequently converted to the t-butyldimethylsilyl ether and t-butyldimethylsilyl ester, and analyzed … Show more

“…The eluate was collected in reaction vials and evaporated to dryness. Glycerol was derivatized to its tbutyldimethylsilyl derivative essentially as described previously (8). Briefly, 50-60 l of anhydrous acetonitrile were added to dried eluates, followed by the addition of 20 l of N-methyl-N-(tertbutyldimethylsilyl)trifluoroacetamide.…”

“…Various problems, analysis methods, and guidelines for the detection of ll-nor-9-carboxytetrahydrocannabinol (THC-COOH) in urine in Japan have been discussed (73). The detection of THC-COOH in urine has been done by SPE, derivatization, and GC/MS (74), liquid/liquid extraction, derivatization, and GC/MS (75), and SPE, derivatization, and GC/MS/MS (76).…”
