Conformational and stacking properties of 3′-5′ and 2′-5′ linked oligoribonucleotides studied by CD

Abstract: Comparative CD studies have been carried out to characterize the properties of 2′–5′ and 3′–5′ oligoriboadenylates and oligoribouridylates from dimer to decamer. The CD band of the 3′–5′ oligoribonucleotides was larger than that of the 2′–5′ oligoribonucleotides and increased with the increase in chain length, while the CD band of the 2′–5′ oligoribonucleotides increased little beyond the dimer level. The CD analysis of the chain length dependency revealed that the 3′–5′ oligoribonucleotides adopt mainly the b… Show more

“…It is now well established that nucleic acids, even with 2¢,5¢ linkages, associate to form Watson and Crick paired duplexes [2][3][4][5]10,11,[32][33][34][35][36][37]. They also selectively associate with DNA and RNA with a varying degree of stability.…”

“…D-and L-adenosine 5 -phosphorimidazolides were prepared from D-and L-adenosine 5 -phosphate, respectively, with imidazole using dipyridyl disulfide and triphenylphosphine as a condensing agent (Lohrmann and Orgel, 1978;Sawai et al, 1989). Authentic samples of 2 -5 and 3 -5 linked D-oligoadenylates were prepared by the methods described previously (Sawai et al, 1996). Authentic samples of diastereomers of adenylyladenosine, ApA, were prepared by Urata et al (Urata et al, 2002).…”

“…The CD bands of 2Ј,5Ј phosphorothioate oligoadenylates decreased with increasing temperature as did those of the corresponding 2Ј,5Ј phosphodiester oligoadenylates. 27 The temperature-dependent change in the CD of these compounds is ascribed to a shifting of the stackedunstacked two-state equilibrium, because isoelliptic points were observed in the CD spectra. The equilibrium constant is expressed by and [] u values at 270 nm in Table I.…”
