Conformational Dynamics of the Trp-Cage Miniprotein at Its Folding Temperature

Abstract: The folding temperature of the trp-cage mini-protein was determined to be in the range 311-317 K depending on the method used. Our study is focused on determining the structure and dynamics of the polypeptide chain close to its unfolding or melting temperature. At T = 305 K, Trp6-Arg16 and Trp6-Pro12 long-range interactions are observed, and at T = 313 K, only the Trp6-Arg16 interactions remain, while all of mentioned interactions are observed in the native state of the protein. Partial (at T = 305 K) and comp… Show more

“…While there are unanswered questions concerning the Trp-cage folding pathway, with a number of groups having reported data suggesting that Trp-cage folding is not a two-state system, 19,28,3335 these data are largely associated with structure melting analysis rather dynamics measurements. Both T-jump and NMR relaxation experiments are consistent with a net two-state process, with fewer exceptions.…”

“…For Trp-Cage, some alternatively folded conformations have even lower enthalpy than the native structure (Figure 1b). In the presence of such conformations, 68 therefore, the conformational space would not be described by a simple two-state model with a folded conformation and an unfolded state of disordered conformations. Cage is not a two-state folder.…”
