Conjugated linoleic acid isomers strongly improve the redox status of bovine mammary epithelial cells (BME-UV1)

Abstract: Some studies have shown the protective effects of conjugated linoleic acid (CLA) isomers against oxidative stress and lipid peroxidation in animal models, but no information is available about CLA and changes in oxidative status of the bovine mammary gland. The objectives of the study were to assess in vitro the effect of CLA on the cellular antioxidant response of bovine mammary cells, to examine whether CLA isomers could play a role in cell protection against the oxidative stress, and to study the molecular … Show more

“…In our study, and in accordance with that previously reported by Szczechowiak et al , the increase in concentration of vaccenic acid could be favoured by the action of bioactive compounds present in the DGP which tend to inhibit the terminal steps of ruminal biohydrogenation, thus avoiding the formation of stearic acid (C18: 0) . The importance of vaccenic acid lies in the fact that this compound represents the substrate of the mammary gland Δ9‐desaturase, which is responsible for the production of at least 90% of rumenic acid, which is reported to have a relevant role in mammary gland functionality, because of its antioxidant activity that preserve bovine mammary epithelial cells from lipoperoxidation . Moreover, CLA are compounds with high biological value for humans, due to their potential activity in slowing the development of atherosclerosis, modulating the immune system and influencing glucose and lipid metabolism .…”

“…Recently, Basiricò et al. (, ) showed an improved antioxidative status in bovine mammary epithelial cells. During the intramammary LPS challenge, plasma concentration of glucose increased in both treatment groups in agreement with earlier studies (Vernay et al., ; Waldron et al., ; Zarrin et al., ).…”

“…Not much research was carried out regarding CLA effects on the immune system of transition dairy cows. Supplementation of bovine mammary epithelial cells with CLA improved their oxidative status (Basiricò, Morera, Dipasquale, Tröscher, & Bernabucci, ; Basiricò et al., ). Dänicke et al.…”

“…Due to the high metabolic rate of the mammary epithelial cells during lactation, it can produce large amounts of ROS and lipid peroxides in vivo, especially in high-yielding dairy cows. If these excessive oxygen free radicals are doi: 10.17221/8851-CJAS not cleared in time, oxidative stress occurs when there is an imbalance between the production of ROS and the host antioxidant capabilities (Basirico et al 2015), and contributes to compromised immunity and increases incidence of diseases (Sordillo and Aitken 2009). Selenoprotein can scavenge hydrogen peroxide, lipid peroxide radicals and phospholipids, and protect tissues against oxidative damage (Bruzelius et al 2008).…”
