Conserved architecture of the core RNA polymerase II initiation complex

Abstract: During transcription initiation at promoters of protein-coding genes, RNA polymerase (Pol) II assembles with TBP, TFIIB and TFIIF into a conserved core initiation complex that recruits additional factors. The core complex stabilizes open DNA and initiates RNA synthesis, and it is conserved in the Pol I and Pol III transcription systems. Here, we derive the domain architecture of the yeast core pol II initiation complex during transcription initiation. The yeast complex resembles the human initiation complex an… Show more

“…The computational models in Figure 6 also imply that p53 bound to Pol II would be in close proximity to the binding site of the core initiation machinery, specifically the GTFs TFIID and TFIIB, (Geiger et al 1996;Nikolov et al 1996;Kostrewa et al 2009;Muhlbacher et al 2014). This observation is supported by several lines of evidence.…”

“…RNA synthesis leads to the initially transcribing complex (ITC) and later to release of the general factors and formation of the elongation complex. Recent studies elucidated the three-dimensional architecture of initiation intermediates, including the ITC [3][4][5][6][7][8][9] .…”

“…cMed corresponds to the endogenous core Mediator purified from yeast 23 , and has a human counterpart that was reported while our manuscript was in revision 24 . cMed binds the core ITC (cITC), which lacks TFIIE and TFIIH and is stabilized by a short six-nucleotide RNA 6,9 . We resolved the architecture of the cITC-cMed complex by cryo-EM and protein crosslinking.…”

“…This includes information about the subunit organization of the polymerases themselves as well as the organization of polymerases with additional proteins into higher-order functional units. Studies have targeted RNA Pol I [81,82], II [83], and III [84] as well as the Pol II-PIC complex [85,86] and the Pol II-capping enzyme complex [87]. A subcomplex of the Mediator complex (the so-called middle module) [88] and the Mediator head module in complex with the C-terminal domain of Pol II [89] were also investigated by XL-MS. More recently, XL-MS was used to provide spatial restraints on the Mediator core and the Pol II-Mediator core initiation complex in the most comprehensive study on polymerases and associated complexes to date [90].…”
