Conserved baculoviral ORFs 10 and 14 from Bombyx mori multiple nucleopolyhedrovirus

Abstract: ABSTRACT. ORFs 10 and 14 from Bombyx mori multiple nucleopolyhedrovirus (BmMNPV) were amplified, cloned and sequenced. Nucleotide analysis of these genes and those of other baculoviruses showed that these genes are highly conserved. The p10 protein from BmMNPV ORF10 has 70 amino acid residues similar to that of the four other known BmNPV strains. The BmMNPV ORF14 alignment showed a higher identity with the nucleopolyhedrovirus ORF14 from the baculovirus BmNPV and from Autographa californica multiple nucleopoly… Show more

“…To con rm whether larvae are positive for virus infection after BmNPV inoculation, the fatty body DNA of the healthy control group and those groups inoculated with BmNPV, untreated and treated with Bm5, were analyzed. The genomic DNA extraction protocol used was according to (Monesi et al 1995) with modi cations (Santos et al 2010). The quanti cation of the DNA concentration was performed using a NanoDrop® ND-1000 UV-Vis, and the 193 bp segment of BmNPV Orf14 (KF306217.1) was ampli ed to prove infection in BmNPV samples.…”

“…Viral DNA was obtained according to the methodology previously described [20]. The DNA was extracted from geographic isolated BmNPV, which was conceded by R. C. Brancalhão and L. F. C. Ribeiro, from Universidade Estadual do Oeste do Paraná (Unioeste).…”

“…These primers were used to amplify the BmNPV DNA. According to Santos et al [20], the ORF 14 codes a protein from NPV viral envelope and it is associated with stress response, such as the ones caused by balucovirus infection in B. mori. The primers ORF14 Forward (5'ATG GAC GGT GTA AAG TTG CTG G3') and ORF14 Reverse (5'TCA AAA TCA ACG CCG TCG TC3') amplify a fragment of 443 base pairs from BmNPV genome.…”

“…Sericulture in Brazil is an activity developed mainly by families on small rural properties, contributing to improving workers' income and reducing rural exodus (Meng et al, 2017). This configures a low technological level for the breeding environment of silkworm, Bombyx mori L. (Lepidoptera: Bombycidae), and among the problems associated with sericulture that directly affect the quality of the threads, the diseases caused by entomopathogenic viruses, bacteria, and fungi stand out (Potrich et al, 2007; Santos et al, 2010; Margatho et al, 2012). The high susceptibility of silkworm caterpillars to these microorganisms is due to the domestication process that the silkworm has gone through (Rajagopal et al, 2014).…”
