Consistent, multi‐instrument single tube quantification of CD20 in antibody bound per cell based on CD4 reference

Abstract: Detecting changes in the expression levels of cell antigens could provide critical information for the diagnosis of many diseases, for example, leukemia, lymphoma, and immunodeficiency diseases, detecting minimal residual disease, monitoring immunotherapies and discovery of meaningful clinical disease markers. One of the most significant challenges in flow cytometry is how to best ensure measurement quality and generate consistent and reproducible inter-laboratory and intra-laboratory results across multiple c… Show more

“…1.29.1) assumes that the labeled antibody against the unknown and labeled anti-CD4 antibody have similar effective fluorescence per antibody values (so called effective F/P). Several studies support the assumption that different antibodies against different antigens with the same fluorophore label can have similar effective F/P values if these antibodies are produced by the same manufacturer (Wang et al, 2006;Degheidy et al, 2014Degheidy et al, , 2015. However, this assumption has not been verified with results of unknown antibodies with the same fluorophore label produced by different manufacturers.…”

“…Once the ABC scales are put in place, users can then quantify test biomarkers relative to the CD4 reference marker using test antibodies (with the same fluorophore labels and high binding affinities and specificities) against the test biomarkers. Three published studies (Wang et al, 2006;Degheidy et al, 2014Degheidy et al, , 2015 have provided evidence supporting the assumption that different antibodies against different antigens with the same fluorophore label can have similar effective fluorescence per antibody values if the antibodies are produced by the same manufacturer.…”

“…Cytometer setup and tracking beads (CST, BD Biosciences) were used for daily quality control and setup of initial PMT voltages. On the first day of experiment, PMT voltage of the PE channel was optimized to ensure the MedFI of unstained cells is approximately 2.5 times above the electron noise of the PE channel according to previously published protocol [45]. QuantiBrite PE beads were then run; the resulting MedFI values of the four PE intensity beads were recorded and used as target values to ensure consistency between experiments performed at different days.…”

“…This is a convenient way to calibrate and track the day-to-day performance of the flow cytometer or to ensure crossinstrument standardization [9,12]. Practical solutions to accomplish standardization have been described e lsewhere [13,14].…”
